摘 要:目的研究miR-132靶向YAP后对肝癌Huh7细胞生长的抑制,为临床治疗肝癌的方法提供新的理论依据。方法实验分为三组:未转染miR-132的Huh7细胞组(control组),阴性对照组(miR NC组)以及转染miR-132的Huh7细胞组(miR-132组),通过RT-PCR检测YAP mRNA表达情况;通过流式细胞仪检测细胞凋亡改变;通过Edu检测细胞增殖能力;通过Transwell法检测细胞侵袭能力。结果 RT-PCR检测YAP mRNA发现miR-132可明显抑制YAP基因在肝癌细胞株Huh7中的表达,control组、miR NC组和miR-132组YAP mRNA的相对表达量为1.000 0±0.024 1、0.942 6±0.024 8和0.257 3±0.045 8(P<0.05);流式细胞仪检测发现转染miR-132后能明显促进肝癌Huh7细胞的凋亡,control组、miR NC组和miR-132组Huh7细胞总凋亡为4.03±0.35、4.53±0.26和13.62±0.49(P<0.05);EdU检测发现转染miR-132后肝癌Huh7细胞增殖能力明显受抑制,control组、miR NC组和miR-132组分别为(65.12±1.93)%、(64.02±0.87)%和(42.00±1.26)%(P<0.05);traswell检测结果发现转染miR-132后肝癌Huh7细胞侵袭能力明显受抑制,control组、miR NC组和miR-132组分别为3.35±0.13、0.72±0.06和0.48±0.03(P<0.05)。结论 miR-132具有抑制肝癌细胞生长的作用,并有可能通过靶向YAP而下调其表达起作用。Objective To research the effect of miR-132 targeted YAP to the hepatocellular carcinoma Huh-7 cell growth and find new theoretical basis for liver hepatocellular carcinoma clinical treatment. Methods The cells were divided into three groups:not transfection of miR-132 group(control group), negative control group(miR NC group), and transfection of miR-132 group(miR-132 group). Cell of the YAP gene expression was detected by the real-time PCR.Cell apoptosis ability of change was detected by the flow cytometry. Cell proliferation of change was detected by the EDU. Made use of Transwell to detect cell invasion ability of the change. Results The real-time PCR detection results showed that hepatocellular carcinoma Huh-7 cell of the YAP gene expression obviously inhibited, control group, miR NC group and miR-132 group relative expression were 1.000 0±0.024 1, 0.942 6±0.024 8 and 0.257 3±0.045 8(P〈0.05).The flow cytometry detection results showed that hepatocellular carcinoma Huh-7 cell apoptosis rate obviously promoted after transfection miR-132, control group, miR NC group and miR-132 group of total apoptosis rate were 4.03±0.35, 4.53±0.26 and 13.62±0.49(P〈0.05). The EDU detection results showed that hepatocellular carcinoma Huh-7 cell proliferation ability obviously inhibited, control group, miR NC group and miR-132 group were(65.12±1.93)%,(64.02±0.87)% and(42.00±1.26)%(P0.05), and Transwell test found that hepatocellular carcinoma Huh-7 cell invasion abilities were restrained, control group, miR NC group and miR-132 group were 3.35±0.13, 0.72±0.06 and 0.48±0.03(P〈0.05). Conclusion MiR-132 has inhibit the growth of hepatocellular carcinoma cells and may have targeted by YAP driving downregulation.
