摘  要:目的观察巨噬细胞活化后小鼠真菌性角膜炎中的细胞因子一氧化氮合酶(iNOS)、巨噬细胞移动抑制因子(MIF)、髓过氧化物酶(MPO)的表达水平,探讨机体免疫在该病发生发展中的作用。方法应用角膜表层镜法建立Balb/c小鼠茄病镰刀菌性角膜炎模型,分别在小鼠角膜感染真菌前注入乳胶微粒和PBS液,形成实验组和对照组。在感染后第1,3,5,7d,裂隙灯显微镜观察各组角膜炎特点;H-E染色观察角膜病理变化;Real timePCR检测角膜中细胞因子MIF、MPO、iNOS的表达。结果 (1)临床特征:实验组,早期角膜浸润混浊且进行性加重,第7d最严重;对照组,早期角膜浸润混浊进行性加重,第5d后新生血管大量生长。(2)H-E染色:实验组可见早期角膜中央大量炎症细胞浸润,至第7d最重。对照组可见早期角膜缘、角膜基质及前房中有炎症细胞浸润,第5d后明显减少,基质纤维细胞逐渐增多。(3)Real time-PCR检测结果表明,MPO、MIF、iNOS细胞因子基因mRNA在接种真菌后第1d,实验组较对照组低,随后实验组细胞因子浓度持续升高,且均高于对照组,于第7d达到顶峰。结论活化角膜巨噬细胞后,MPO、MIF、iNOS等细胞因子在真菌性角膜炎中的表达增强,可能在真菌性角膜炎的病理生理中起重要作用。Objective To detect the expression of cytokine of iNOS,MIF,MPO in a mouse model of Fusarium solani keratitis after macrophage activation,and investigate the role of immune response in this disease.Methods A mouse model of Fusarium solani keratitis was established by epikeratophakia.Macrophage was activated by intracorneal instillation of latex beads in Balb/c mice before inoculation.On day 1,3,5,7 after inoculation,the appearance of corneal ulcer was observed by slitlamp,infected corneas were subjected to histopathological analysis,cytokine mRNA levels of iNOS,MIF,MPO were measured by real-time polymerase chain reaction (PCR).Results In the Latex Beads-treated group,it showed heavy inflammation,diffuse opacity and resulting in corneal perforation mostly on day 7 after inoculation.By contrast,the control group showed much neovascularization on 5 days after inoculation.Histopathologic examination of cornea showed,much more marked dense inflammatory cell infiltration in the Latex Beads-treated group than control group.Expression of iNOS,MIF,MPO mRNA was some lower in the Latex Beads-treated group on day 1 after inoculation,but were significantly enhanced versus control group,and reached a peak at 7 day after inoculation.Conclusions In fungal keratitis,local activation of macrophages may upregulate the expression of cytokine of iNOS,MIF,MPO,which may play an impor tant role in the pathophysiology of fungal keratitis.
