胃癌脂质变化的傅立叶变换红外光谱研究及细胞膜磷脂组分变化的分析

摘　　要：目的观察胃癌中脂质变化的傅立叶变换红外光谱(FTIR)改变及细胞膜磷脂组分的变化。方法应用FTIR分别对胃癌及癌旁组织、胃癌细胞株、胃癌及癌旁组织的细胞膜进行检测,对胃癌及癌旁组织细胞膜运用高效液相色谱法(HPLC)进行分析。结果 FTIR检测结果显示脂类相关谱带强度在癌旁组织中强于癌组织,而在组织细胞膜中的强度癌组织大于癌旁组织;HPLC分析结果显示磷脂酰肌醇(PI)、磷脂酰乙醇胺(PE)、磷脂酰胆碱(PC)3种膜磷脂含量在胃癌中增加且差异具有统计学意义(P<0.01),磷脂酰丝氨酸(PS)改变虽无显著性意义,但在胃癌组织细胞膜中含量也高于癌旁组织细胞膜。结论胃癌与癌旁组织相比脂类含量减少,但癌组织细胞膜的脂类含量高于癌旁组织细胞膜,胃癌发生过程中磷脂代谢发生了改变,对其脂类变化的深入研究对探讨胃癌的发生、发展,寻找新的治疗靶点和判断临床预后具有重要意义。Objective To observe lipid change in gastric cancer by FTIR and to analyze gastric cancer cell membrane phospholipid variation. Methods FTIR was applied to study gastric cancer tissue and para-cancer tissue, gastric cancer cell line and gastric cancer tissue cell membrane and para-cancer tissue cell membrane, respectively. Gastric cancer tissue cell membrane and para-cancer tissue cell membrane were analyzed by HPLC. Results The FTIR study result displayed the intensity of lipids related bands are stronger in para-cancer tissue than those in gastric cancer tissue but weaker in para-eancer tissue cell membrane than those in gastric cancer tissue cell membrane. The analytic result of HPLC showed the content of membrane phospholipid PI, PE and PC increased in the gastric cancer tissue cell membrane and have significant statistical deviation （P^0.01）. The content of membrane phospholipid PS is more in gastric cancer tissue eel[ membrane than in para-cancer tissue cell membrane but do not have significant statistical deviation. Conclusion The content of lipids decreased in gastric cancer tissue compared with para-cancer tissue. But membrane phospholipid increased in gastric cancer tissue cell membrane compared with para-cancer tissue cell membrane; phospholipid metabolism changed in the carcinogenesis of gastric cancer. Further study in the variation of lipids will have significance in the carcinogenesis and development of gastric cancer, in the search of new therapy target, and in the judgment of clinical prognosis.
