RIN1过表达对人胃腺癌MKN28细胞增殖、侵袭、迁移和凋亡的影响

摘　　要：[目的]研究RIN1过表达对人胃腺癌MKN28细胞增殖、侵袭、迁移和凋亡的影响。[方法]①采用Western blot法和免疫荧光法检测RIN1在MKN28细胞中表达;②采用Burd-U标记检测MKN28细胞高表达RIN1后细胞增殖能力变化;③采用Transwell法、细胞划痕法检测MKN28细胞高表达RIN1后细胞侵袭和迁移能力变化;④采用AV/PI染色法、Hoechst染色法检测MKN28细胞高表达RIN1后细胞凋亡情况。[结果]①RIN1在MKN28细胞中呈高表达,表达量是空载对照组的3.22倍,且主要分布在细胞质中;②Burd-U染色结果显示,RIN1高表达MKN28细胞的阳性率显著性升高,与空载对照组相比为75.6%±5.4%vs 25.6%±1.1%(P<0.01);③RIN1高表达MKN28细胞的侵袭和迁移能力明显增强,与空载对照组相比分别为122.0±7.0 vs 65.00±2.52(P<0.01)和54.22%±3.45%vs 38.6%±2.45%(P<0.01);④RIN1高表达MKN28细胞株细胞凋亡数显著性减少,与空载对照组相比为21.63%±2.60%vs 8.07%±1.60%(P<0.01)。[结论]RIN1过表达促进MKN28细胞增殖、侵袭和迁移,抑制MKN28细胞凋亡;RIN1可能是介导胃癌发生的癌基因之一。[ Purpose] To investigate the effect of RIN 1 overexpression on proliferation, invasion, migration and apoptnsis of human gastric a（tenncarcinoma cell line MKN28. [Methods] RIN1 expression in pcDNA3.1（＋）/ RIN1/ MKN28 cells were identified by Western blot and Immunofluorescence. The effect of RINI overex- pression on proliferatinn,invasion,migration and apoptosis of MKN28 cells were analyzed by Brd-U assay, Transwell chamber and Scratch Wound Healing, AV/PI dye method and Hoechst staining, respectively. [Re- suits 1 R1N1 was highly expressed in pcDNA3.1 ,（＋）/RIN I/MKN28 cells. The quantity of RINI expression on pcDNA3.1 （＋）/RIN I/MKN28 cells was 3.22 times of pcDNA3.1 （＋）/MKN28 cells, and RIN 1 mainly dis- tributed in the cytoplasm. Compared with pcDNA3.1（＋）/MKN28 cells,the proliferation ability of pcDNA3.1 （＋）/RIN 1/M KN28 （＇ells （75.6%±5.4% vs 25.6%± 1.1%, P〈0.01） was significantly enhanced, and the migrated eell number （122.0±7.0 vs 65.00＋2.52,P〈0.01） and the migrated rate of pcDNA3.1 （＋）/RINI/MKN28 cells （54.22%±3.45% vs 38.6%±2.45%,P〈0.01） were markedly increased and the apoptotic rate of pcDNA3.1 （＋）/ RIN I/MKN28 cells （8.07%±1.60% vs 21.63%±2.60% ,P〈0.01） was dramatically reduced.[Conclusion] RIN overexpressed in MKN28 （,ells is eapable of prnmoting proliferation,invasion,migration and inhibiting apop- tosis.and RIN1 nmy be one of mwogenes to take part in the occurrence of gastric cancer.
