摘 要:目的:探讨CHFR(checkpoint with FHA and ring finger)基因启动子异常甲基化在宫颈癌发生发展过程中的作用及其在宫颈癌早期诊断中的价值。方法:用甲基化特异性PCR方法检测42例宫颈癌组织、30例宫颈上皮内瘤样病变组织(cervical intraepithelial neoplasia,CIN)及20例正常宫颈组织中CHFR基因启动子区CpG岛甲基化状态。结果:宫颈癌组织及CIN中CHFR基因异常甲基化检出率分别为33.3%(14/42)和6.7%(2/30),正常宫颈组织中未检出CHFR基因甲基化;三组间CHFR基因异常甲基化发生率之间比较差异具有统计学意义(χ2=10.69,P=0.005);CHFR基因甲基化发生率与年龄、病理分级、临床分期之间差异无统计学意义(P>0.05)。结论:CHFR基因的异常甲基化参与了宫颈癌的发生和发展过程,其甲基化的检测对宫颈癌的早期诊断有一定的价值。Objective To investigate the role of CHFR gene promoter hypermethylation in the development of cervical cancer and its value in early diagnosis. Methods The hypermethylation of CHFR gene promoter CpG islands were detected in 42cancer tissues,30 CIN tissues and 20 normal cervical tissues by methylation specific PCR,respectively. Results The positive rate of CHFR gene promoter hypermethylation was 33. 3%( 14 /42) in cervical cancer,6. 7%( 2 /30) in CIN tissues,no hypermethylation was found in normal tissues. The positive rate among three different tissues was significant difference( χ2=10. 69,P = 0. 005),but no different in age,pathological grade,clinical stage of patients,respectively( P〈0. 05). Conclusion The hypermethylation of CHFR gene could involove in the development of cervical cancer,which provided certain value in early diagnosis.
