摘　　要：目的探讨MiR-145表达升高对口腔鳞状细胞癌细胞CAL27生物学特性的影响。方法通过实时定量PCR检测MiR-145在正常口腔黏膜细胞hNOK和口腔癌细胞CAL27的表达水平。通过MiR-145对CAL27的细胞转染,CAL27集落形成实验、细胞周期与凋亡实验,观察CAL27细胞生物学特性的改变。结果与正常口腔黏膜细胞hNOK相比,口腔癌细胞CAL27中的MiR-145表达呈下调状态。通过恢复口腔癌细胞中MiR-145的表达显著抑制了CAL27细胞集落的形成,并诱导G1期阻滞和细胞凋亡。结论 MiR-145可发挥口腔细胞癌的抑制功能,抑制口腔癌细胞CAL27细胞增殖特性,诱导其凋亡过程。在其诊断与治疗应用中,恢复MiR-145表达可能是一种合理的方法。Objective To explore the effects of up-regulated MiR-145 expression on the biological characteristics of human oral carcinoma cell line CAL27.Methods By real-time quantitative PCR,we examined the level of MiR-145 expression in normal oral mucous membrane cell hNOK and oral carcinoma cell CAL27.Through CAL27 cell transfection by MiR-145,CAL27 colony formation test,cell cycle and cell apoptosis experiment,we observed the changes in the biological characteristics of CAL27.Results Compared with that of normal oral mucous membrane cell hNOK,the expression level of oral carcinoma cell CAL27 MiR-145 was obviously down-regulated.Improving the expression of oral carcinoma cell CAL27 MiR-145 could obviously inhibit CAL27 colony formation and induce the arrest of G1 phase cell cycle and cell apoptosis.Conclusion MiR-145 can exert the function of restraining oral cell carcinoma through inhibiting CAL27 cell proliferation and inducing the apoptosis process.During diagnosis and treatment of oral cell carcinoma,improving MiR-145 expression may be a reasonable method.
