摘 要:目的研究微波热疗对提高阿霉素(doxorubicin,DOX)体外抑制人RB Y79细胞的作用。方法分别使用MTT法、荧光显微镜、流式细胞仪检测人RB Y79细胞的存活率、摄取及凋亡情况;使用分光光度法检测Y79细胞内Caspase-3的活性。将实验分为空白对照组、DOX组、微波热疗组、DOX+微波热疗组,分别将4组进行以上细胞实验。结果DOX+微波热疗组与其他组相比细胞存活率明显下降,差异均有统计学意义(均为P<0.05)。细胞荧光摄取实验表明,DOX+微波热疗组与DOX组相比,其细胞穿透效果在60min内呈明显上升趋势。DOX+微波热疗组72 h细胞凋亡率可达(49.08±1.02)%,可有效促进细胞凋亡,与其他各组相比差异有明显统计学意义(均为P<0.01)。同时,DOX组、微波热疗组、DOX+微波热疗组、空白对照组人RB Y79细胞的Caspase-3活性分别为2.88±0.16、1.61±0.09、4.67±0.27、1.00±0.05,微波热疗+DOX可以有效提升细胞内Caspase-3活性,与其余各组间差异均有统计学意义(均为P<0.05)。结论微波热疗可以有效提高人RB Y79细胞体外化学治疗的效果,此疗法是一种很有前景的治疗视网膜母细胞瘤的方法。Objective To study the ability of microwave hyperthermia in enhancing chemotherapeutic efficacy of doxorubicin (DOX) against human retinoblastoma Y79 cells in vitro. Methods MTT method was used to detect the viability of Y79 cells, cellular uptake of Y79 cells was determined by fluorescence microscopy, cell apoptosis was detected by flow cytometry, and spectrophotometric method was used to detect the expression of Caspase-3 in Y79 cells. The experiments were divided into 4 groups:blank control group, DOX group, microwave treatment group and DOX combined with microwave treatment group, and these groups were experimented, respectively.Results The Y79 cells exposed to DOX with microwave treatment showed a significant decrease in cell viability which compared with other groups ( all P 〈 0.05 ). The cellular uptake results showed that the cells increased permeability to DOX immedi- ately with the addition of microwave compared with DOX alone, which continued to increase over 50 minutes. Apoptotic rate of cells treated with DOX and microwave hyperthermia was (49.08 ± 1.02 ) % , and it could enhance the apoptosis rate compared to other experimental groups ( all P 〈 0.01 ). Meanwhile, the activity of Caspase-3 in cells of blank control group, DOX group, microwave treatment group and DOX combined with microwave treatment group were 2.88 ±0.15,1.51 ± 0.09,4.57 ± 0.27 and 1.00 ± 0. 05, respectively, DOX with microwave treatment could enhance the activity of Caspase-3 in cells, there were statistical differences compared with other groups ( all P 〈 0.05 ). Conclusion Microwave hyperthermia can enhance the chemotherapeutic efficacy against retinoblastoma Y79 cells in vitro, DOX combined with microwave treat- ment is a promising approach for retinoblastoma.
