棒柄花苷A对照品的制备研究

摘　　要：【目的】建立从棒柄花(Cleidion brevipetiolatumPaxet Hoffm)中制备棒柄花苷A(PPGA)对照品的方法。【方法】利用硅胶柱层析、重结晶和反相制备高效液相色谱对棒柄花的乙醇提取物进行分离纯化,以HPLC对棒柄花苷A进行纯度检查和含量测定,并通过UV,IR,MS,1 HNMR等对其进行结构确证。【结果】从棒柄花叶中分离、纯化出棒柄花苷A对照品,质量分数W>98.0%。【结论】所建立的制备方法简单,制备出的棒柄花苷A对照品符合中药化学对照品的相关要求,可作为棒柄花药材和含棒柄花成药质量控制,以及中药药效物质基础用的化学对照品。[Objective]A seperation method was established to prepare reference substance of PP-GA from leave of Cleidion brevipetiolatum Paxet Hoffm.[Methods]Ethanol-extract of the leave of Cleidion brevipetiolatum Paxet Hoffm was isolated and purified by RP-HPLC combi-ning with column chromatography and recrystallization.Its structure was identified by UV,IR, MS,1 HNMR.[Results]PPGA was completely seperated from leave ofCleidion brevipetiolatum Paxet Hoffm.The purity of the reference substance was not less than 98.0%.[Conclusion]The method is simple and convenient,by which the prepared compound is accorded with the relative standards of chemical reference substance in Chinese materia medica.It can be used as a refer-ence subsatance for the quality control and the research of herbal medicine.
