摘 要:目的研究大肠癌患者外周血自然杀伤(NK)细胞的数量和活性的变化及意义。方法用流式细胞仪分析检测25例大肠癌、60例大肠息肉、30名健康者的外周血NK细胞的数量,并用酶标仪检测各组NK细胞的细胞毒活性。结果大肠癌患者的NK细胞数、NK细胞杀伤率[(1.1±0.7)×108、(26±7)%]均明显低于正常组[(2.8±1.3)×108、(66±9)%]、大肠息肉组[(2.6±1.2)×108、(62±7)%],差异均有统计学意义(均P<0.05)。结论大肠癌患者外周血NK细胞数量及活性明显降低,提高大肠癌患者NK细胞的数量及活性可增强大肠癌的细胞过继免疫治疗。Objective To research the changes and significance of NK cells number and cytotoxicity from peripheral blood in patients with colorectal carcinoma. Methods Flow cytometry was used to determine the number of NK cells from peripheral blood in patients with colorectal carcinoma (25 cases), colorectal polyps (60 cases), and healthy adults (30 cases). The microplate reader was used to detect cytotoxicity of NK cells in all cases. Results Between the number ofNK cells(× 108) and the killing rate of NK cell for K562 cell, the colorectal carcinoma group [(1.1 ± 0.7) × 108, (26 ± 7)%] decreased significantly as compared with the healthy group [(2.8 ± 1.3) × 108, (66 ± 9)%] and the colorectal polyps group [(2.6 ± 1.2) × 108, (62 ± 7)%] (P 〈0.05). Conclusions The number and cytotoxicity of NK cells decreases significantly from peripheral blood in patients with colorectal carcinoma. Enhancing the number and cytotoxicity of NK cell in patients with colorectal carcinoma can strengthen adoptive cellular immunotherapy of colorectal carcinoma.
