摘 要:目的探讨非小细胞肺癌(Non-SmallCellLung Cancer,NSCLC)组织中Raf激酶抑制蛋白(RafKinase Inhibitor Protein,RKIP)和磷酸化细胞外信号调节激酶(phosphorylation of Extracelluar signal-Regulated Kinase,p-ERK)的表达及其与临床病理特征的关系。方法应用免疫组织化学方法检测80例NSCLC组织及相应的癌旁正常肺组织中RKIP与p-ERK蛋白的表达情况,分析两者与NSCLC临床病理学参数的关系。结果 NSCLC组织中RKIP的表达低于癌旁正常肺组织,而p-ERK的表达高于癌旁正常肺组织,差异有统计学意义。其表达水平与NSCLC的分化程度、TNM分期、淋巴结转移及术后生存时间有明显关系;RKIP蛋白与p-ERK蛋白的表达水平呈负相关(P<0.05)。结论 RKIP与p-ERK的表达在NSCLC的发生、发展中起重要作用,RKIP可能是NSCLC的一个转移抑制基因,RKIP表达下调导致p-ERK的过度表达从而促进肺癌细胞的侵袭和转移。Objective To investigate the expression of Raf kinase inhibitor protein (RKIP) and phosphorylation of extracelluar signal-regulated kinase (p-ERK) in non-small cell lung cancer (NSCLC) and to explore its correlation with the clinicopathologic features. Methods The expression of RKIP and p-ERK was detected using immunohistochemistry in 80 cases of NSCLC specimens and 80 cases of adjacent normal lung tissue. Statistical analysis was used to determine the relationship between their expressions and clinicopatbological parameters. Results Statistical analysis revealed that RKIP positive staining rate was lower in the tumor tissues than that in normal lung tissues. But p-ERK positive staining rate was significantly higher in tumor tissues than that in normal lung tissues (P〈0.05). The expression of RKIP and p-ERK was significantly related with the different differentiation of tumor, TNM stage, lymph node metastasis and the survival time (P〈0.05, respectively). There was a negative correlation between the expression of RKIP and p-ERK in NSCLC (P〈0.05). Conclusion The expression of RKIP and p-ERK may play an important role in the initiation and development of NSCLC. RKIP may be a metastasis suppressor of NSCLC, and decreased RKIP expression is associated with the increased metastasis of lung carcinoma cells through the over-expression of p-ERK.
