复方排毒止带汤含药血清对人宫颈鳞癌SiHa细胞人乳头瘤病毒16型E6,E7,p53,Rb蛋白表达的影响

摘　　要：目的采用蛋白免疫印迹法(Western Blot)研究中药复方排毒止带汤含药血清对人宫颈鳞癌SiHa细胞人乳头瘤病毒16型(HPV16)E6,E7,p53,pRb蛋白表达的影响。探讨复方排毒止带汤对人乳头瘤病毒的影响。方法将大鼠随机分为空白对照组、25%含药血清组及50%含药血清组。空白对照组口服生理盐水,25%含药血清组及50%含药血清组口服复方排毒止带汤煎剂,将复方排毒止带汤25%含药血清和50%含药血清分别作用于SiHa细胞72,120 h后,采用Western Blot法检测HPV16 E6,E7,p53,pRb蛋白表达,采集数据。统计学分析采用独立样本t检验。结果复方排毒止带汤50%含药血清作用72 h,HPV16 E6,E7蛋白表达水平下降,p53,pRb蛋白表达水平升高,复方排毒止带汤50%含药血清作用120 h,HPV16 E6,E7蛋白表达水平较作用72 h明显下降,p53,Rb蛋白表达水平明显升高,2组相应指标比较P均<0.05,差异有统计学意义。复方排毒止带汤25%含药血清作用120 h,HPV16 E6,E7蛋白表达水平下降,p53,Rb蛋白表达水平上升,复方排毒止带汤50%含药血清作用120 h HPV16 E6,E7蛋白表达水平较25%含药血清明显下降,p53,Rb蛋白表达水平明显升高,2组相应指标比较P均<0.05,差异有统计学意义。结论复方排毒止带汤含药血清对HPV16 E6,E7,p53,Rb蛋白表达具有明显调控作用,复方排毒止带汤的活性具有一定的时效性。Objective To study the effect medicated serum of Compound Detoxification Zhidai Decoction on protein expres- sion of human papilloma virus type 16 E6, E7, p53, Rb in human cervical carcinoma SiHa cells by Western Blot method and explore the effect of Compound Detoxification Zhidai Decoction on human papilloma virus. Methods The rats were random- ly divided into control group, 25% of the drug-containing serum group and 50% of the drug-containing serum group. The control group was given oral saline solution,while 250/0 of the drug-containing serum group and 50% of the drug-containing serum group were given Compound Detoxification Zhidai Decoction. 25% drug-containing serum and 50% drug-containing serum were used to applied to SiHa cells. After 72 h and 120 h, we adopted Wsetern Blot method to detect protein expres-sion. Statistical analysis was performed using independent sample t test. Results HPV16 E6, E7 protein expression level decreased and p53 and pRb protein expression level rised after 72 h with 50% drag-containing serum. But HPV16 E6, E7 protein expression level obviously decreased and p53 and pRb protein expression level obviously rised compared with those of 72h with 50% drug-containing serum after 120 h with 50% drug-containing serum. There was a significant difference be- tween the two groups. There was a significant difference between the two groups （P 〈 0.05）. HPV16 E6, E7 protein expres- sion level decreased and p53 and pRb protein expression level rised after 120h with 25% drug-containing serum. HPV16 E6, E7 protein expression level obviously decreased and p53 and pRb protein expression level obviously rised compared with those of 120 h with 25% drug-containing serum after 120 h with 50% drug-containing serum. There was a significant differ- ence between the two groups （ P 〈 0.05）. Conclusion The effect of medicated serum of Compound Detoxification Zhidai Decoction on protein expression of HPV16 E6, E7, p53, Rb in human cervical carcinoma SiHa cells is obvious.
