摘  要:目的:探索不同超声参数对肿瘤细胞生长的影响,为超声联合载药微泡肿瘤治疗研究奠定实验基础。方法:采用正交设计的方法,将3种超声参数即超声强度(1.48W/cm2,2.56W/cm2,2.79W/cm2)、占空比(1∶5,1∶1,5∶1,超声波发出时间与间歇时间之比)和超声时间(5mins,10mins,15mins)按3因素3水平分成9组,分别作用于SKOV3和MCF-7肿瘤细胞株,并设空白对照组(不作任何处理),每种肿瘤细胞各10组。采用MTT法测定两种细胞生长情况,并对这3种超声参数进行析因分析。结果:与空白对照组相比,占空比为1∶5和1∶1时,两种肿瘤细胞生长均未见明显抑制(P>0.05);占空比达到5∶1时,3组SKOV3细胞的生长抑制率达到90%以上(P<0.05),而3组MCF-7细胞的生长抑制率分别为48.8%、55.4%和88.7%(P<0.05)。析因分析结果显示,这3种超声参数均有交互作用,对SKOV3和MCF-7肿瘤细胞生长均有抑制作用,其中占空比是最主要的影响因素,其次为超声强度。结论:不同超声参数对肿瘤细胞生长的抑制作用不同,而占空比是体外超声抑制肿瘤细胞生长的最主要影响因素。Objective: To explore the effect of different ultrasound parameters on tumor cell growth and provide the experimental data for ultrasound combined with microbubbles loaded drug in tumor therapy. Methods: Bases on three factors of the ultrasonic intensity( 1.48W/cm2,2.56W/cm2,2.79W/era2 ) , duty cycle ( 1: 5,1: 1,5:1 ) and ul trasonic exposure time (5mins, 10rains, 15rains), nine groups were designed by orthogonal methods in the SKOV3 cell line and MCF 7 cell line with different ultrasonic treatment as well as the control group (without any treatment)was designed. Their growth inhibition rate was evaluated by MTT assay and these three parameters were in turn analyzed. Results:Compared with the control group, the cellular growth inhibition were not observed in two cell lines at the duty cycles of 1 : 1 and 1 : 5. However, the cellular growth inhibition rates were more than 90% ( P 〈 0.05 ) in three groups of SKOV3 cell line at the duty cycle of 5:1 while it was 48.8% ,55.4% and 88.7% in three groups of MCF 7 cell line respectively ( P 〈 0.05 ). Factorial analysis showed three ultrasonic parameters had interaction effect on the cellu lar viability in SKOV3 cell line and MCF 7 cell line, but the duty cycle of the ultrasound was the most important fac tor. Conclusion:The role of different ultrasound parameters on tumor cells growth inhibition is different, but the duty cycle of the ultrasound was the most important factor in the effect of the uhrasound on tumor cells growth inhibition in vitro.
