摘  要:目的:观察共培养DC-LAK细胞生物学特性及经不同途径输注体内后的组织器官分布特点。方法:以荧光染料CFSE标记共培养DC-LAK细胞,后经瘤旁皮下注射或腹腔注射途径输注于荷LLC肿瘤C57BL/6小鼠体内,使用激光共聚焦荧光显微镜检测共培养DC-LAK细胞在各器官内的动态分布。结果:共培养DC-LAK细胞输入荷瘤小鼠体内后,腹腔注射途径中腹腔内器官率先达到分布高峰。瘤旁皮下注射途径中肿瘤率先达到分布高峰;两种途径中肿瘤内的分布DC-LAK组均高于LAK组。结论:DC与LAK细胞共培养可增加LAK细胞对肿瘤的靶向性,瘤旁注射途径可提高肿瘤内的分布,体腔输入途径更适合于体腔内恶性渗液或局限病灶的治疗。Objective:To observe the biological characteristics of co - culture DC - LAK cells and the distribution of co - culture DC - LAK cells after different pathway infusion in tumor - bearing mice. Methods: Co - culture DE - LAK cells labeled by CFSE had been infusioned in the tumor - bearing mice. Laser scanning confocal microscope (LSM) was used to detect the distribution. Results:The DC - LAK cells after intraperitoneal injection in tumor - bearing mice were major accumulated in liver and spleen. After para - tumor subcutaneous injection in tumor - bearing mice, DC - LAK cells were major accumulated in tumor tissue. Both tow pathway, DC co - culture with LAK ceils can raise the accumulation of LAK cells in tumor tissue. The distribution of I)C - LAK cells of para - tumor subcutaneous injection group was more than intraperitoneal injection group all the time. Conclusion: The co -culture of DCs and LAK ceils can addable the target to tumor. After para - tumor subcutaneous injection in tumor - bearing mice, DC - LAK ceils are major accumulated in tumor tissue and application by means of body - cavity way should suit for malig- nant efusions and limited lesion in it.
