摘　　要：目的:研究膀胱移行细胞癌组织中NY-ESO-1的表达并探讨其临床意义。方法:收集70例膀胱移行细胞癌患者癌组织(Ta-T1期48例,T2-T4期22例;G1级30例,G2级22例,G3级18例)及其中10例患者癌旁正常组织,采用反转录聚合酶链反应(RT-PCR)技术检测NY-ESO-1 mRNA的表达;采用Western blot技术检测上述组织中NY-ESO-1蛋白的表达。结果:NY-ESO-1 mRNA在70例膀胱移行细胞癌组织中28例(40.0%)表达阳性,10例癌旁组织表达均阴性;NY-ESO-1蛋白在22例(31.4%)中表达阳性,10例癌旁组织表达均阴性。NY-ESO-1 mRNA和蛋白的表达均和膀胱癌的病理分级相关,有统计学差异(P<0.05),其表达率随着肿瘤分级的增高而升高。NY-ESO-1 mRNA和蛋白在不同膀胱癌分期中的表达无显著性差异(P>0.05)。结论:NY-ESO-1在膀胱移行细胞癌组织中有较高表达,其表达率随着肿瘤分级的增高而升高,而在癌旁组织无表达,从而有望成为膀胱移行细胞癌特异性免疫治疗的靶分子及判断预后的指标。Objective：To explore the expression profile of NY - ESO - 1 in transitional cell carcinoma of the blad- der,and investigate its possible correlation with clinicopathologic characteristics, nethods：NY -ESO - 1 mRNA ex- pression was detected by reverse transcription polymerase chain reaction （RT- PCR） in 70 cases of transitional cell carcinoma of the bladder（Ta - Tm 48 cases, T2 - T4 22 cases ; G1 30 cases, G2 22 cases, G3 18 cases） as well as 10 ca- ses of adjacent normal bladder mucosa. NY - ESO - 1 protein expression was detected by Western blot. Results： Of 70 cases of transitional cell carcinoma of the bladder,NY - ESO - 1 mRNA was detected in 28 cases（40.0% ） and NY - ESO - 1 protein was detected in 22 cases（31.4% ）. There was a significant correlation between NY - ESO - 1 ex- pression at both mRNA and protein levels with higher grades of the disease （P 〈 0.05 ）. Conclusion： NY - ESO - 1 is highly expressed in transitional cell carcinoma of the bladder, and therefore may serve as a therapeutic target of specif- ic immunotherapy as well as a potential prognostic marker for transitional cell carcinoma of the bladder.
