ING4在卵巢肿瘤中的表达及其临床意义

摘　　要：目的:研究ING4(inhibitor of growth family member 4)在卵巢肿瘤中的表达,探讨其与卵巢癌发生发展及预后的关系。方法:应用免疫组化SP法检测104例卵巢组织中ING4的表达情况,RT-PCR检测80例卵巢组织ING4 mRNA表达水平。结果:免疫组化:正常卵巢组织、卵巢交界性肿瘤组织、卵巢癌组织中ING4蛋白表达阶梯性降低,三组间差异有统计学意义(P<0.001)。ING4蛋白的表达随着FIGO分期升高和组织学分级的降低表达减弱,各组间有显著差异(P<0.05)。随着ING4蛋白表达减弱,患者生存时间减少。RT-PCR:ING4 mRNA在正常卵巢组织中表达高于卵巢癌组织,有统计学意义(P<0.05)。ING4 mRNA的表达与患者的临床分期显著相关(P<0.05)。结论:ING4蛋白与卵巢癌的发生、发展和转移具有相关性,在一定程度上可判断卵巢癌恶性程度和评估临床预后,同时为卵巢癌的靶向治疗提供一个新的靶点。Objective：To study expression and clinical significance of ING4 gene in epithelial ovarian cancer. Methods ：Expression of ING4 gene was evaluated by immunohistochemistry method in 104 cases and reverse tran- scription polymerse chain reaction（ RT -PCR） in 80 cases. Survival probabilities were calculated by the product limit method of Kaplan - Meier. Univariate and multivariate Cox regression models were performed for analysis. Results： Positive expression rate of ING4 in tumor tissues was significantly stronger than in borderline tissue of epithelial ovari- an tumor and normal ovarian tissues. In a multivariate Cox regression model, positive expression of ING4 gene, clinical stage, histological grade,lymph node metastasis and histological types were associated with overall survival. Conclu- sion ：ING4 gene participates in the occurrence,invasion and metastasis of ovarian cancer,which can be used as an in- dex to judge the malignant degree of ovarian cancer and evaluate prognosis to some extent.
