摘 要:以大真眼点藻(Eustigmatos magnus)为研究材料,改良BG-11作为基础培养基,设置四种NaNO3浓度,利用O3 cm的玻璃柱状光生物反应器对该藻进行培养,通过一系列生理指标的测定来探讨不同NaNO3浓度对大真眼点藻生长和脂积累的影响。实验结果表明:最适宜该藻生物量积累的NaNO3浓度为9 mmol·L–1,此条件下生物量达到11.31 g·L–1;通过脂组分分级,结果显示3.6 mmol·L–1实验组的总脂含量和中性脂含量均为最高,总脂含量为60.21%(干重百分比),中性脂含量为92.99%(总脂百分比);主要脂肪酸组分为棕榈酸、棕榈油酸和油酸,三者含量总和占总脂肪酸含量的55%-87%;9 mmol·L–1实验组的总脂单位体积产率最高为0.29 g·L–1·d–1,6 mmol·L-1实验组的中性脂产率最高,达到0.26 g·L–1·d–1。总体来看,9 mmol·L–1实验组和6 mmol·L–1实验组的产油能力相当,但若要进行工业化生产,则6 mmol·L–1为产油脂的最适浓度。The effects of sodium nitrate concentration on the growth and the physiological characteristics of Eustigmatos magnus, which was cultured in the column photobioreactors (3 cm in diameter) with the BG-11 medium were investigated. The results showed that the optimum sodium nitrate concentration for the growth of E. magnus was 9 mmol·L-1 and the highest biomass was 11.31 g·L-1. Neutral lipid, glycolipid and phospholipid were partitioned from total lipid of this alga through C 18 cartridge. Under the sodium nitrate concentration of 3.6 mmol·L-1, the total lipid content reached 60.21%(% dry weight) and the neutral lipid content reached 92.99%(% total lipid), respectively. Both of the values were higher than the other three groups. In addition, the main components of fatty acid were palmic acid, palmitoleic acid and oleic acid, while the sum of their contents achieved 55% to 87% (% total fatty acid). The optimal concentration of sodium nitrate for the volumetric productivity of total lipid was 9 mmol·L-1 and the value was up to 0.29 g·L-1·d-l, meanwhile the maximum volumetric productivity of neutral lipid ran up to 0.26 g.L-Ld-I under the concentration of 6 mmol·L-1. Considering the cost of sodium nitrate, 6 mmol·L-1 is the most economical concentration for commercial production.
