灭幽汤对幽门螺杆菌相关性胃炎小鼠胃黏膜组织TLR2、NF-κBp65的影响

摘　　要：目的探讨灭幽汤对幽门螺杆菌相关性胃炎(HAG)小鼠胃黏膜组织Toll样受体2(TLR2)、核转因子p65(NF-κBp65)的影响。方法将BALB/c小鼠随机分为正常对照组、模型组、胃三联组、灭幽汤组。采用复合因素(肥甘食物+湿热环境+幽门螺杆菌)建立BALB/c小鼠HP相关性胃炎脾胃湿热证模型。采用免疫组化法检测胃黏膜组织TLR2、NF-κBp65的蛋白表达情况。结果小鼠模型组与正常对照组比较TLR2与NF-κBp65表达均增加(P<0.05);与模型组比较,灭幽汤组小鼠胃黏膜TLR2与NF-κBp65表达均降低(P<0.05)。结论灭幽汤能根除HP,可减轻HP引起的胃黏膜炎症反应。其机制可能是通过下调TLR2,抑制NF-κBp65的激活,从而阻止NF-κB的活化,减轻胃粘膜炎症反应,进而发挥其治疗作用。Objective To explore effects and action mechanism of Mieyou Decoction on TLR2 and NF-K BP65 in gastric mucosa tissues of mice with Helicobacter pylori-related gastritis Methods BALB/c mice were randomly assigned to normal control group, model group, gastric triad group, Mieyou group, developed a model of HAG BALB/c mice Spleen-stomach Damp-heat Syndrome by composite factors （high sugar and fat food ＋ hot and humid environment ＋ H. Pylori）. The immunohistochemical assay was adopted to detect the protein expressions of nuclear transcription factor TLR2 and NF-κBP65. Results Compared with normal control group, the expressions of TLR2 and NF-κBp65 in gastric mucosa of mice （model group） increased （P〈 0.05）, compared with model control group, the expressions decreased in Mieyou Decoction group （P〈0.05）. Conclusion Mieyou Decoction can eradicate H. pylori, regulate the expressions of TLR2 and NF-κBp65 in gastric mucosa of mice, alleviate gastric mucosal inflammation. HAG was treated by Mieyou Decoction probably through down-regulating the expressions of TLR2, inhibiting the activation of NF-κBp65, thus hindering the activation of NF-κB, then producing treatment effect.
