大蒜素类化合物局部电子云密度对活性氧生成的影响

摘　　要：合成新的大蒜素类化合物(DATSs),研究这类化合物对佛波酯PMA刺激巨噬细胞生成的活性氧(超氧阴离子O-·2,过氧化氢H2O2,羟基自由基OH·)可能产生的影响.利用半经验方法计算DATSs的结构参数,分析这些参数与其影响活性氧产生的关系.研究发现,在一定范围内,DATSs的C(2)带负电荷越多则可诱导PMA刺激MΦ产生更多的O-·2;增加C(1)的负电荷或减少C(3)的负电荷均能够诱导H2O2的生成增多;当DATSs的脂溶性接近时,C(2)所带负电荷越多,则MΦ生成的H2O2越多;DATS-1,DATS-2,DATS-3和DATS-5对PMA刺激MΦ产生OH·有抑制作用.表明DATSs局部电子云密度与佛波酯PMA刺激巨噬细胞活性氧的生成密切相关,这些化合物可能通过抑制OH·发挥抗氧化作用.The novel diallyl trisulfide analogues have been synthesized and assayed for the possible effect on the production of reactive oxygen species including superoxide anion (O-· 2 ),hydrogen peroxide (H2 O2 ) and hydroxyl radical (OH ·)in phorbol myristate acetate stimulated peritoneal macrophages.A set of molecular properties of diallyl trisulfide and its analogues (DATSs)have been analyzed via the semiempiri-cal method,to compare with the results of activity assays.The results show that more negative charge at C(2) induced PMA to stimulate MФto produce higher O-· 2 count.More amount of H2 O2 has induced by in-creasing the negative charge at C(1)or decreasing the negative charge at C(3),more H2 O2 has been given by MФwith more negative charge at C(2)when the DATSs has similar hydrophobicity.The release of OH· in MФhas been reduced by treatment with DATS-1,DATS-2,DATS-3 and DATS-5.It is suggested that the partial electron density at DATSs may be one of the important factor that effects the production of ROS from PMA stimulated MФ.And those compounds exhibited antioxidantive activity might through in-hibiting the production of OH·.
