二甲双胍对糖尿病大鼠微种植体支抗骨结合的影响

摘  要:目的二甲双胍干预治疗糖尿病大鼠,观察其对微种植体支抗骨整合的影响。方法30只Wistar大鼠,6~8周龄雄性,随机抽取10只作为正常组,其余20只采用高脂肪饲料加链脲佐菌素(STZ溶液40mg/kg),诱导建立2型糖尿病大鼠模型。建模成功后,随机分为模型组、药物组。三组大鼠于左侧胫骨各植入微种植体2枚,药物组应用二甲双胍灌胃治疗[剂量0.2g/(kg·d)],2周后加力60g,12周处死,测血糖、血胰岛素、总胆固醇、甘油三酯、并计算胰岛素敏感指数。制作含有微种植体的组织学切片,观察并计算微种植体骨结合率及骨矿化沉积率。结果三组大鼠微种植体仅有模型组1枚微种植体脱落。用药组的骨结合率及骨矿化沉积率高于模型组(P〈0.05)。结论二甲双胍有控制血糖、调节骨代谢的作用,可提高微种植体的骨结合,但作用有限。Objective To observe the influence of metformin intervention to microimplant osseointegra- tion of diabetic rats. Methods Thirty Wistar rats(6 - 8 weeks, male) were feed, 10 rats were randomly selected as normal group, 20 rats were establish the Ⅱ diabetic rats model by highly fat diet and streptozotocin(STZ 40mg/kg). After that, all rats were divided in model group and drug group randomly. Three groups implanted two microimplants into left tibia, drug group was treated by infusing the stomach with the metformin, 0.2g/(kg·d) , burned 60g after 2 weeks and rats were sacrificed after 12 weeks, blood glucose, blood insulin, total cholesterol, triglyceride and calculating insulin sensitivity were measured. The microimplant bone contacted ratio and bone mineral apposition rate (MAR) were calculated by making the histological sections contain micro - implant. Results Only one of all micro - implant was abscisiced in model group. The bone contact ratio and bone mineral apposition rate I MAR) in drug group were higher than that in model group(P〈0.05). Conclusion Metformin can cnotrol blood glucose, adjust the remolding of bone and improve micro-implant osseointegration, but the action is limit.
