Wnt-1、uPA在子宫内膜癌中的表达及其相关性的研究

摘　　要：目的探讨Wnt信号传导通路在子宫内膜癌侵袭、转移中作用机制,并结合uPA蛋白在子宫内膜癌中的表达并分析二者之间的相关性。方法采用RT-PCR技术,对10例手术切除的新鲜子宫内膜癌和手术切除的新鲜子宫内膜组织进行Wnt-1和uPA mRNA的检测;采用免疫组化SP法对89例子宫内膜癌和30例正常子宫内膜组织中Wnt-1和uPA蛋白的表达进行检测。结果 Wnt-1的高表达与肿瘤的手术分期和淋巴结转移相关(P<0.05);uPA与手术分期、肌层浸润和淋巴结转移相关,具有统计学意义(P<0.05),且Wnt-1在子宫内膜癌组织中阳性表达与uPA的高表达之间呈正相关关系(r=0.434,P<0.05)。结论 Wnt-1和uPA在子宫内膜癌中的表达存在着相关性,推测Wnt-1异常表达通过下游uPA的过度表达促进子宫内膜癌细胞黏附性下降,间质降解和转移。Objective To study the mechanism of invasion and metastasis of Wnt signal transduction pathway, and analyze the correlation of uPA and Wnt- 1 protein expression in endometrial carcinoma. Methods Totally 10 cases of fresh endometrial cancer and endometrial tissue by surgical resection were collected, and then Wnt - 1 and uPA mRNA were detected with RT - PCR. The expression of Wnt- 1 and uPA protein was detected on 89 endometrial cancers and 30 normal endometrial tissues with immunohistochemical SP method. Results The high expression of Wnt - 1 was associated with surgical stages of tumor and lymph node metastasis（P〈0.05）. The uPA was associated with surgical stages, myometrial invasion and lymph node metastasis（ P〈0.05）, there was statistical significance. The positive expression of Wnt- 1 in endometrial carcinoma was positive correlation to the high expression of uPA. （r = 0. 434,P〈0.05）. Condusions The expression of Wnt- 1 and uPA is positive correlation in endometrial carcinoma. The abnormal expression of Wnt- 1 can decrease adhesion, degrade interstitial cell and transferring of endometrial carcinoma through the overexpression of the downstream uPA.
