摘  要:DNA甲基化是一种表观遗传修饰,抑癌基因高甲基化与多种肿瘤的发生有关。目前化疗是治疗肿瘤最主要的手段,而化疗耐药是化疗失败和肿瘤复发的重要原因之一。多数肿瘤中存在抑癌基因高甲基化的情况,这与化疗耐药的形成关系密切。去甲基化药物可以激活抑癌基因启动子并恢复肿瘤细胞对化疗药物的敏感性,逆转肿瘤对化疗药物耐药性。在常见的异常甲基化基因中已证实去甲基化可逆转化疗耐药。DNA methylation is a kind of epigenetic modification,hypermethylation of antioncogene is closely related the occurrence of many kinds of tumor.At present,chemotherapy is a main method for the treatment of tumor.However,chemotherapy resistance is one of the most important reasons for tumor recurrence and the failure of chemotherapy.Tumor suppressor genes are methylated in the majority of the tumor,which is closely related to the chemotherapy resistance.Demethylation drugs can activate tumor suppressor gene promoter and restore the sensitivity of tumor cells to the chemotherapy drugs,or even reverse the chemotherapy resistance.It has been confirmed that demethylation can reverse the drug resistance in the common abnormal methylated genes.
