摘  要:目的观察普罗布考对血管性痴呆大鼠认知功能及海马区bcl-2、bax蛋白表达的影响。方法选取健康雄性SD大鼠,采用双侧颈总动脉永久性结扎法建立血管性痴呆大鼠模型,随机分为假手术组、模型组、普罗布考组,每组12只。给药8 w后,采用Morris水迷宫实验,对各组大鼠进行行为学测试。用western blot方法和免疫组织化学方法测定海马区bcl-2和bax蛋白的表达变化。结果与假手术组相比,模型组大鼠逃避潜伏期时间延长,穿越平台次数减少,海马区bcl-2蛋白表达明显下降,bax蛋白表达明显上升;给予普罗布考后,与模型组相比,普罗布考组大鼠逃避潜伏期时间缩短,穿越平台次数增多,海马区bcl-2蛋白表达明显上升,bax蛋白表达明显下降。结论普罗布考可以改善血管性痴呆大鼠学习记忆能力,其机制可能与上调bcl-2蛋白表达,下调bax蛋白表达有关。Objective To observe the effect of probucol on vascular dementia rats in ethology and expression of bcl- 2 and bax protein. Methods Healthy male SD rats were used to establish vascular dementia rat model by permanent oc- clusion of bilateral common carotid artery. The rats were randomly divided into sham operated group, model group,probucol group, and there were 12 rats in each group. Morris water maze was used to evaluate learning and memory ability of rats in each group after 8 weeks administration. After ethology test, the expression of bcl-2 and bax protein in hippocampus were examined by western blot and immunohistochemical staining. Results Compared with sham operated group ,the escape la- tency of model group rats was significantly prolonged, and the frequency of crossing platform reduced, the expression level of bcl-2 in hippocampus in model group was significantly lower than sham operated group, but higher for the expression level of bax;Compared with model group,the escape latency of probueol group rats was shorted,and the frequency of crossing plat- form increased,the expression level of bcl-2 in hippocampus in probucol group was significantly higher than model group, but lower for the expression level of bax. Conclusions Probueol can improve learning and memory ability of vascular de- mentia rats.and the mechanism may be related to increase bcl-2 expression and decrease bax expression.
