摘 要:目的观察氟对大鼠肝组织caspase-3和caspase-9基因表达的影响,探讨其对肝组织损伤的作用机制。方法将48只SD雄性大鼠随机分为:对照组、低氟组、中氟组和高氟组,每组12只,分别饮用含0,50,100,200 mg/L氟化钠去离子水,采用自由饮水方式进行连续染毒120 d。测定肝系数;采用荧光定量PCR检测肝组织中caspase-3、caspase-9基因表达水平。结果染氟组与对照组相比,体重差异无统计学意义。染氟组大鼠肝脏脏器系数与对照组相比差异无统计学意义。低、中、高染氟组caspase-3基因表达量分别是对照组的0.34±0.10、1.37±0.43和2.27±0.73倍;低、中、高染氟组caspase-9基因表达量分别是对照组的1.03±0.36、3.05±0.68和5.85±0.52倍。结论氟化物对肝细胞DNA损伤以及诱导细胞的凋亡,可能是通过激活caspase-3和caspase-9而促进了它们的发生。Objective To investigate the effects of fluoride on expression of caspase-3 and caspase-9 gene in the liver tissues of rats,and explore the effect and mechanisms of fluoride on injury of hepatic tissues.Methods Forty-eight male SD rats were randomly divided into control group(n =12),low-dose fluorine group(n =12),middle-dose fluorine group(n =12) and high-dose fluorine group (n =1 2).SD rats were administered with 0,5 0,1 0 0,and 2 0 0 mg/ L sodium fluoride for 1 2 0 d,respectively.The liver coefficient was determined,and the expression levels of caspase-3 and caspase-9 mRNA in liver tissues were detected by real-time PCR.Results There was no significant difference in body weight and liver coefficient between fluoride exposed groups and control group.The expression of caspase-3 in low-,middle-and high-dose fluorine groups was 0.34 ± 0.10,1.37 ± 0.43 and 2.27 ± 0.73 times the amount of control group.The expression of caspase-9 in in low-,middle-and high-dose fluorine groups was 1.03 ± 0.36,3.05 ± 0.68 and 5.85 ± 0.52 times the amount of control group.Conclusion Fluoride can cause DNA damage and induce cell apoptosis probably through activating the expression of caspase-3 and caspase-9.
