加味导赤散与维生素B_(12)联合灌胃对大鼠复发性口腔溃疡模型外周血T淋巴细胞亚群的影响

摘　　要：目的探讨加味导赤散与维生素B12对大鼠复发性口腔溃疡(ROU)模型外周血T淋巴细胞的影响及治疗作用。方法免疫法建立大鼠ROU动物模型,异体大鼠口腔黏膜匀浆组织加完全弗氏佐剂作为抗原,注射在脊柱两侧皮内,每2周注射一次,共注射5次。将实验大鼠随机分为4组:正常组、溃疡组、左旋咪唑组和加味导赤散+维生素B12组。灌胃20 d后流式细胞术检测各组大鼠外周血T淋巴细胞亚群CD4+、CD8+细胞数量及CD4+/CD8+。结果实验大鼠于第3次注射1周后开始出现口腔溃疡。溃疡组大鼠外周血CD4+细胞数量CD4+/CD8+较正常组降低(P<0.05),CD8+改变无统计学意义(P>0.05)。实验大鼠给药后,加味导赤散+维生素B12组和左旋咪唑组外周血CD4+细胞数量、CD4+/CD8+均有回升,较溃疡组差异有统计学意义(P<0.05)。两组之间比较差异没有统计学意义(P>0.05)。结论 ROU的发生与T淋巴细胞失调有关,加味导赤散与维生素B12能改善ROU大鼠外周血T淋巴细胞亚群失衡。Objective To explore the effect of Chinese herb plus vitamin B12 on peripheral blood T-lymphocyte subsets in recurrent oral ulceration（ROU） rats.Methods Experimental animal model of ROU was established by injecting antigens into two sides of SD rat＇ s spinal column.The antigens were made by mixing Freund＇ s complete adjuvant（FCA） with allogenic rat oral mucosa homogenate.The rats were randomly divided into 4 groups：normal group,model group,levamisole group and Chinese herb plus vitamin B12 group.The levels of CD4 ＋,CD8 ＋ in blood were observed using flow cytometry at 20 d after intragastric administration.Results ①The ulcers were found at one week after the third injection.②Compared with normal group,the levels of CD4 ＋ and CD4 ＋/CD8 ＋ decreased in model group（P ＜ 0.05）,while there was no significant change in CD8＋ （P ＞ 0.05）.③Compared with model group,the levels of CD4 ＋ and CD4 ＋/CD8 ＋ increased in levamisole group and Chinese herb plus vitamin B12 group （P ＜ 0.05）,but there was no significant difference between the two groups （P ＞ 0.05）.Conclusion The occurrence of ROU is related to the imbalance of T-lymphocyte subsets.Chinese herb plus vitamin B12 may treat the ROU rats by improving the imbalance of T-lymphocyte subsets.
