摘 要:采用乙醇-硫酸铵双水相体系分离纯化棉花叶中总黄酮,确定其双水相体系组成为23%无水乙醇-22%(NH4)2SO4,通过单因素实验和Box-Behnken Design实验研究粗提液质量分数、pH、NaCl质量分数三个变量对棉花叶中总黄酮响应值的影响程度,用响应面法得出三个考察因素最优工艺参数,即:粗提液质量分数31%、pH4、NaCl质量分数3%,在此条件下萃取率为76.32%。与响应面法优化黄酮萃取率预测值78.02%接近,为棉花叶中总黄酮有效开发利用提供一定的借鉴作用。An aqueous two-phase system composed of 23% ethanol and 22% (NH4)2SO4 was used to extract total flavonoids from cotton leaves. Mass fraction of crude extract, pH and mass fraction of NaCI on the extraction efficiency of total flavonoids were explored by one-factor-at-a-time and Box-Benhnken design methods. The results showed that the optimal conditions were as follows :crude extract concentration 31%, pH4, NaCI concentration 3%. Under the above conditions the confirmed tests indicated that the extraction rate of total flavonoids was 76.32% which had close relation with the predicted yield of the extraction rate of total flavonoids was 78.02%. The study would provide reference for the effective utilization of fiavonoids from cotton leaves.
