苯丁酸钠对体外人肺腺癌细胞A549的影响

摘　　要：目的观察苯丁酸钠(PB)对体外人肺腺癌细胞A549的影响。方法采用MTT法观察不同浓度PB对人肺腺癌细胞A549增殖的抑制作用,流式细胞术分析不同浓度PB对生长期人肺腺癌细胞A549的细胞周期及凋亡情况的影响。结果 PB作用后人肺腺癌细胞A549的生长缓慢,细胞阻滞于G0/G1期(P<0.05),凋亡增加(P<0.05),其抑制作用呈时间和浓度依赖性。结论 PB能抑制人肺腺癌细胞A549的生长,促进其凋亡。Objective To investigate the effects of sodium phenylbutyrate（ PB） on human lung adinocarcinoma cell A549. Methods The inhibition effects of PB on the proliferation of human lung adinocarcinoma cell A549 was assayed by MTT method in different concentration. The alteration of cell cycle phase and apoptosis were detected by using the fluorescence flow cytomtry. Results After the treatment of PB,the growth of human lung adinocarcinoma cell A549 was lower,most of the cells were arrested in G0/G1 phase,and the cell apoptosis was increased（ P 〈0. 05）. The effects showed time- and dose-dependent pattern. Conclusion The growth of the lung adenocarcinoma A549 cells can be inhibited by PB in vitro. The mechanism may be increase of G0/G1 fraction and induction of apoptosis.
