鲜食山楂新品种蒙山红组织培养初探

摘　　要：为获得适宜山楂蒙山红快速繁殖的最佳培养基组合,以山楂新品种蒙山红和大金星的当年生带芽茎段为外植体,设置8种培养基组合进行愈伤组织诱导和芽增殖培养,分析两个山楂品种的组织培养的最适培养条件。结果表明:蒙山红诱导愈伤组织的最适培养基是MS+KT 1.0mg·L-1+IBA 0.1mg·L-1,最适宜的芽诱导培养基是MS+KT 1.0mg·L-1+IBA 0.5mg·L-1。大金星诱导愈伤组织的最适培养基是MS+KT0.5mg·L-1+IBA 0.5mg·L-1,最适宜的芽诱导培养基是MS+KT 1.5mg·L-1+IBA 0.5mg·L-1。全部培养基均附加蔗糖30g·L-1,琼脂7g·L-1,pH5.8。In order to obtain the most appropriate rapid propagation medium of hawthorn variety of Mengshanhong,taking stems section from Mengshanhong and Dajinxing as explants. Eight kinds of medium combination on callus induction and bud multiplication culture were set up to analyze the optimum culture conditions of two hawthorn varieties. The results showed that the best culture medium for callus induction was MS ＋KT 1.0 mg·L-1＋IBA 0.1 mg·L-1 for Mengshanhong,and the best culture medium for bud proliferation was MS＋ KT 1.0 mg·L-1 ＋IBA 0.5 mg·L-1. The best culture medium for callus induction was MS＋KT 0.5 mg·L-1 ＋ IBA 0.5 mg·L-1 for Dajinxing,and the best culture medium for bud proliferation was MS＋KT 1.5 mg·L-1 ＋ IBA 0.5 mg·L-1. 30 g·L-1 sugar and 7 g·L-1 agar were added to all culture medium,and pH was 5.8.
