摘 要:目的:研究刺山柑果实中硫代葡萄糖苷酶解的影响因素,确定硫苷酶解的最佳条件并分析和鉴定酶解产物。方法:以刺山柑果实粉末为材料,研究不同的温度、pH以及时间等单因素试验和正交试验筛选优化水解硫代葡萄糖苷的最佳条件,并且利用气相色谱-质谱连用技术分析鉴定了酶解产物。结果:酶解时间为140 min,水解温度为50℃,pH值为7.0是最佳酶解条件。利用气相色谱-质谱连用技术分析鉴定出3种异硫氰酸酯化合物,占硫苷酶解产物中挥发性化合物总量的80%。结论:确定了刺山柑果实中硫代葡萄糖苷酶解的最佳条件,为刺山柑果实的研究提供了一定的理论基础。OBJECtIVE To research the impact of factors of enzymatic hydrolysis for glucosinolate in Capparis spinosa fruit; to determine the optimum conditions of enzymatic hydrolysis and identify the enzymatic hydrolysis products. METHODS Taking the Capparis spinosa fruit powder as the material, the different temperature, pH and time of single factor test and or- thogonal test were studied to optimize the best conditions of the hydrolysis of glucosinolate; and the enzymatic hydrolysis prod- ucts were identified by GC-MS. RESULTS The results showed that the optimum enzymatic hydrolysis conditions for the glu- cosinolate were as follows:enzymatic time 140 min, temperature 50℃ pH 7. 0, and three isothiocyanate compounds were identi- fied from the enzymatic hydrolysis products of the glucosinolate by GC-MS and they accounted for 80% of the whole volatile products. CONCLUSION This paper identified the optimum conditions of the enzymatic hydrolysis for glucosinolate in Cappa- ris spinosa fruit and provided the theoretical basis of study on Capparis spinosa fruit.
