摘  要:目的探讨在中国北方地区汉族人群中ECE1上rs212528和rs213045两个位点基因分型并分析其多态性与缺血性卒中发病风险的相关性。方法采用病例-对照研究,共选取381名缺血性卒中患者和366名健康对照个体,采用Hardy-Weinberg遗传平衡定律检验基因型分布情况,通过二元Logistic回归进行ECE1变体与缺血性卒中风险的关联分析。结果rs213045 TG基因型与rs213045 GG基因型相比,患缺血性卒中的风险显著增加(P0.03,校正OR1.47,95%CI1.042.07)。在显性模型下,rs213045 TT/TG基因型与rs213045 GG相比,患缺血性卒中的风险显著增加(P0.029,校正OR1.43,95%CI1.041.99)。在rs212528中,CC、CT以及CC/CT基因型与TT基因型相比,患缺血性卒中的风险差异不显著。结论ECE1基因多态性可能与中国北方地区人群缺血性卒中的遗传易感性有关。Objective The study was conducted to genotype the rs212528 and rs213045 loci on ECE1 in Han populations in northern China and analyze the correlation between the gene polymorphism and the risk of ischemic stroke. Methods 381 patients with isehemic stroke and 366 healthy controls were selected and were analyzed for their distribution of genotypes by the Hardy-Weinberg law. Binary logistic regression was used to determine the correlation between ECE1 variants and the risk of ischemic stroke. Results The rs213045 TG genotype had a significantly higher risk of isehemie stroke than the rs213045 GG genotype (P=-0.03, calibrated OR=1.47, 95% CI=1.04-2.07). In dominant model, the risk of ischemie stroke of the rs213045 Tr/TG genotype was significantly higher than the rs213045 GG genotype (P=-0.029, calibrated OR=1.43, 95% CI=1.04-1.99). In rs212528, the differences in the risk of isehemie stroke between CC, CT, CC/CT, and T'F genotypes were not statistically significant. Conclusion ECE1 gene polymorphism may correlate with genetic susceptibility to ischemic stroke among Han populations in northern China.
