摘  要:目的观察有氧运动对肥胖大鼠脂肪组织锌α糖蛋白(ZAG)基因表达的影响及其与胰岛素敏感指数(ISI)的相关性,为肥胖及相关疾病的防治提供新思路。方法对饮食诱导的肥胖大鼠进行6周有氧运动干预,干预后检测大鼠体重增加值、内脏脂肪垫相对重量、空腹血糖(FBG)、血清甘油三酯(TG)、空腹血清胰岛素(FINS)含量及ISI,采用RT-PCR检测脂肪组织中ZAGmRNA表达水平。结果 (1)运动干预后,肥胖运动组(DIO-E)的体重增加值显著低于对照组(P0.01),与肥胖安静组(DIO-NE)相比虽无显著性差异,但呈下降趋势(P0.061)。(2)DIO-E组大鼠内脏脂肪垫相对重量、FBG,、FINS、TG含量显著低于DIO-NE组;而ZAGmRNA表达水平、ISI则高于对照组和DIO-NE组(P0.05,P0.01)。(3)大鼠脂肪组织ZAGmRNA表达水平和体重增加值、内脏脂肪垫相对重量、FBG、FINS呈显著性负相关(P0.05),与ISI呈高度正相关(r0.712,P0.01),和TG无显著相关(P0.05)。结论运动可以上调肥胖大鼠脂肪组织ZAGmRNA表达;ZAGmRNA表达增加可促进机体脂质代谢,提高胰岛素敏感性。Objective This study aimed to observe the impact of aerobic exercise on the expression of Zinc-α 2-glycoprotein (ZAG) mRNA in adipose tissue of high-fat diet (HFD) -induced obese rats and its correlation with ISI. Methods HFD-induced obese rats were administrated with 6-week aerobic exercise. Weight gain, relative weight of visceral fat, FBG, FINS, TG and ISI were measured. The expression of ZAG mRNA in adipose tissue was determined by RT-PCR. Results (1) Weight gain of the DIO-E group was significantly slower than that of control group. Weight gain of the DIO-E group was lower than that of the DIO-NE group, although the difference had no statistical significance (P=-0.061). (2) Following exercise, the relative weight of visceral fat, FBG, FINS, TG significantly decreased (P〈0.05), whereas ZAG mRNA expression in adipose tissue and ISI increased (P〈0.01). (3) Further statistical analysis demonstrated that the expression of ZAG mRNA was negatively correlated with weight gain, relative weight of visceral fat, FBG, FINS (P〈0.05) but positively correlated with ISI (r=0.712, P〈0.01). Conclusion Aerobic exercise improves the lipid metabolism and insulin sensitivity by up-regulating ZAG mRNA expression in adipose tissue of HFD-induced obese rats.
