摘 要:目的探讨中药补肾益脑片抗衰老的分子机制。方法使用DDRT-PCR方法观察中药补肾益脑片对SAMR 10鼠脑组织mRNA表达的影响。对SAMR10小鼠脑组织基因表达进行比较研究。结果发现了2个受药物影响表达上调和6个受药物影响表达下调的基因片段。测序分析结果显示,上述片段其分别与脑脂肪酸结合蛋白7 cDNA、DNA拓扑异构酶I mRNA、小鼠16S核糖体RNA基因、小鼠细胞色素c氧化酶亚单位ⅡmRNA、小鼠磷酸甘油激酶1 mRNA、小鼠蛋白磷酸激酶2A同功催化α亚单位、小鼠细胞色素c、小鼠组织蛋白酶S mRNA序列同源。它们参与了细胞间相互作用、细胞能量代谢、自由基产生、蛋白质合成、DNA合成与修复等过程。其中,多数与衰老直接相关。结论补肾益脑片在分子水平上以多基因、多靶点、多效应方式发挥其抗衰老作用。Objective It is to approach the anti -senescence molecular mechanism of the traditional Chinese medicine Bushen Yinao Tablets .Methods The effect of Bushen Yinao Tablets on mRNA expression of Senescence -Accelerated Mouse P10 (SAMP10) cerebral tissue by DDRT -PCR, and the gene expression of SAMP10 was compared.Results Two up-ex-pression and six down -expression gene fractions were cloned from the TCM given group .They were homologous to the Mus musculus brain fatty acid binding protein 7 (bfabp) cDNA, Mus musculus topoisomerase DNA I (Topo1) mRNA, Mus mus-culus 16S ribosomal RNA gene , Mus musculus cytochrome c oxidase subunit II ( Cox2 ) mRNA, Mus musculus similar to phosphoglycerate kinase 1 mRNA, Mus musculus protein phosphatase 2a catalytic subunit alpha isoform , Mus musculus cyto-chrome c cDNA , and Mus musculus mRNA for cathepsin S .Those genes were involved in cell -cell interaction , cellular ener-gy metabolism , free radical generation , protein synthesis , and DNA synthesis and repair , most of which were involved in se-nescence .Conclusion Bushen Yinao Tablets exert its effect with multiple gene , multiple targets and multiple effects on anti -senescence in molecular level .
