摘  要:目的观察片仔癀对局灶性脑梗死大鼠炎性损伤相关因子肿瘤坏死因子-α(TNF-α)、细胞间黏附分子-1(ICAM-1)及血管细胞黏附分子-1(VCAM-1)含量的影响。方法将74只SD标准性大鼠随机分成7组,即假手术组10只,按照改良的EZ Longa法制备大脑中动脉栓塞大鼠模型64只,将手术后符合评分标准的大鼠分为模型组10只,片仔癀大剂量组10只、中剂量组12只、小剂量组10只,脑得生组和尼莫地平组各10只。造模前给药3 d,造模后给药4 d,处死取材,参照酶联免疫吸附法(ELISA)检测ICAM-1、VCAM-1及TNF-α的含量。结果模型组血清ICAM-1、VCAM-1及TNF-α含量较假手术组升高(P<0.01);各给药组较模型组均有不同程度的降低,其中片仔癀大剂量组、脑得生组及尼莫地平组血清ICAM-1、VCAM-1、TNF-α含量较模型组降低(P<0.01)。结论片仔癀胶囊可以降低局灶性脑梗死后大鼠ICAM-1、VCAM-1及TNF-α的含量,对于神经功能具有保护作用。Objective To observe the effect of Pien Tze Huan on the serum amount of TNF -α, ICAM -1 and VCAM-1 after MCAO operation. Methods Middle cerebral artery occlusion (MCAO) rat model was estab-lished by improved EZ Longa method. 74 healthy mature male SD rats were randomly divided into model group, sham operation group, smalldose, middose, highdose Pien Tze Huan group, positive control group 1 ( nimodipine group) and positive control group 2 ( Naodesheng group). Rats were pretreated for 3 days before modeling and ad- ministered for 4 days after modeling, then were sacrificed. The amount changes of ICAM-1, VCAM-1 and TNF -α were observed by methods of ELISA. Results The amount of ICAM - 1, VCAM-1 and TNF - αin the model group increased obviously after the MCAO operation compared with that in sham operation group ( P 〈 0. 01 ). The expression in each treatment group was lower than that in the model group (P 〈 O. 05 or P 〈 O. 01 ). Conclusion Pien Tze Huan can decrease the serum amount of ICAM -1, VCAM -1 and TNF -αso as to protect the nerve function.
