摘  要:目的 :探讨小鼠正常肠黏膜组织与大肠癌组织β-catenin、E-cadherin和Ki67表达情况及其关系。方法:采用氧化偶氮甲烷诱导建立小鼠大肠癌模型;运用SP免疫组织化学方法检测20只正常小鼠肠黏膜组织与20只小鼠大肠癌组织中β-catenin、E-cadherin和Ki67的表达情况,并分析三者表达的相关性。结果:β-catenin在小鼠大肠癌组织中的异常表达率(70%)较正常小鼠肠黏膜组织(5%)显著升高(P<0.001);E-cadherin在小鼠大肠癌组织中的阳性率(45%)较正常小鼠肠黏膜组织(95%)显著降低(P<0.001);Ki67在小鼠大肠癌组织中的阳性率(80%)较正常小鼠肠黏膜组织(15%)显著升高(P<0.001)。小鼠大肠癌组织中Ki67的阳性表达与β-catenin的异常表达呈正相关(P<0.001),与E-cadherin的阳性表达呈负相关(P<0.001)。结论:E-cadherin低表达、Ki67高表达以及β-catenin异常表达可能在大肠癌的发生发展和转移过程中发挥着重要作用。Objective:To investigate the relationship of β-catenin,E-cadherin and Ki67 expressions between normal mucosa tissue and colorectal cancer tissues in chemically induced mice.Methods:Azoxymethane was used to induce mice and establish the model of colorectal cancer; SP immunohistochemical method was used to detect β-catenin,E-cadherin and Ki67 expressions in normal mucosa tissue and colorectal cancer tissues in mice.Results:The proportions of abnormal expression of β-catenin were 70% in colorectal cancer group and 5% in control group(P<0.001).The positive expression rates of E-cadherin were 45% in colorectal cancer group and 95% in control group (P<0.001).The positive expression rates of Ki67 were 80% in colorectal cancer group and 15% in control group (P<0.001).Moreover,there was a positive correlation between the β-catenin and Ki67 expressions (P<0.001),as well as a negative correlation between the E-cadherin and Ki67 expressions (P<0.001) in colorectal cancer group.Conclusion:Abnormal expressions of β-catenin,E-cadherin and Ki67 may be associated with the growth and invasion of the colorectal cancer and can be used as a marker of progression for colorectal cancer.
