eIF4E和整合素αvβ6在结肠癌组织中的表达及临床意义

摘　　要：目的:探讨e IF4E和整合素αvβ6在结肠癌组织中的表达及临床意义。方法:采用免疫组化SP法检测69例结肠癌及30例正常结肠组织中e IF4E和整合素αvβ6蛋白的表达,并分析二者与结肠癌临床病理因素的关系。结果:结肠癌组织中e IF4E和整合素αvβ6蛋白的表达明显高于正常结肠组织,差异有统计学意义(P<0.01)。e IF4E表达与淋巴结转移有关(P=0.013),整合素αvβ6表达与肿瘤浸润深度、淋巴结转移及临床TNM分期有关(P=0.027,P=0.034,P=0.031)。结肠癌组织中e IF4E与整合素αvβ6蛋白的表达呈低度正相关(P=0.016,r=0.288)。Kaplan-Meier法生存分析显示,e IF4E和整合素αvβ6蛋白的表达与患者5年生存率显著相关(P=0.008,P=0.040)。Cox回归分析显示,e IF4E及整合素αVβ6蛋白的表达水平、组织分化程度、肿瘤浸润深度和年龄是结肠癌的独立危险因素,其中e IF4E高表达和αVβ6阳性表达的相对危险度分别是4.212(95%CI:1.779,9.973)和2.774(95%CI:1.148,6.700)。结论:结肠癌组织中e IF4E和整合素αvβ6蛋白可作为结肠癌患者术后预后不良的独立预测指标,二者潜在的信号通路可能成为治疗结肠癌的新靶点。Objective：To investigate the expression of elF4E and Integrin α v β 6,their clinical significance as well as two proteins＇ correlation in colonic carcinoma tissue.Methods：The expression of elF4E and integrin α v β 6 was detected in 69 colonic carcinoma tissues and 30 normal colon tissues via SP immunohistochemistry method.And the relationship between the two proteins＇expression and clinicopathological characteristics were analyzed.Results：The expression of elF4E and integrin αv β 6 in colon cancer tissue was significantly higher than that in normal colon tissue （P＜0.001）.The expression of elF4E was significantly associated with tumor metastasis,while integrin α v β 6 was related to invasion depth,tumor metastasis and clinical TNM stage （P=0.027,P=0.034,P=0.031 respectively）.Moderately weak correlation exists between the two proteins （P=0.016,P=0.288）.The survival analysis by Kaplan-Meier and Cox regression model showed that protein expression of high eIF4E and positive integrin αv β 6,as independent prognostic factors （RR=4.212,RR=2.774 respectively）,tended to have a significantly poorer 5-year survival rate（P=0.008,P=0.040 respectively）.Conclusion：elF4 E and integrin αv β 6 can be indicators of poor prognosis for patients with colon cancer.And the potential signaling loop involving them may provide a helpful therapeutic target in prevention and treatment of colon cancer.
