几种中药单体促进阿霉素内化协同诱导白血病细胞凋亡

摘　　要：为了分析3种中药单体对白血病细胞内化阿霉素和诱导细胞凋亡的影响,采用体外培养白血病细胞,以10μmol/L阿霉素处理,或同时加入中药单体(去甲基斑蝥素,β-榄香烯和姜黄素)处理细胞。流式细胞术分析处理1,2 h和4 h后细胞内化阿霉素的相对含量。继续培养至16 h,流式细胞术分析细胞凋亡,并用免疫印迹技术分析细胞内活化Caspase-3的表达。结果3种中药单体中,去甲基斑蝥素和姜黄素具有显著促进阿霉素内化和促进细胞凋亡的效应;β-榄香烯也可增强阿霉素诱导细胞凋亡的效应,但对阿霉素内化无显著影响;去甲基斑蝥素和姜黄素与阿霉素联合可增加活化Caspase-3水平。表明3种中药单体均可增强阿霉素诱导白血病细胞凋亡的效应,其中去甲基斑蝥素和姜黄素的增敏效应与其促进阿霉素的内化相关。Objective：To analyze the effect of three kinds of Chinese herbs monomer on the intermalization of adriamycin in leukemia cell and the induction of cell apoptosis.Methods：The leukemia cells were cultured in the medium of 10 μmol/L adriamycin invitro,or the Chinese herbs monomer（norcantharidin,NCTD;β-elemi ene and curcumin） were added.The relative contents of Intra.cellular adriamycin after culturing 1,2 and 4hrs were analyzed with flow cytometry.The cell apoptosis was analyzed till culturing16 hrs with flow cytometry and the expression of Caspase-3 Intracellular activated was analyzed with immunoblotting technique.Re.sults：Among three kinds of Chinese herbs monomer,NCTD and curcumin had significant effects on promoting the intermalization of adriamycin and the induction of cell apoptosis;β-elemi ene enhanced the induction of cell apoptosis by adriamycin and had insignificant effect on the internalization of adriamycin;Three level of Caspase-3 could be enhanced by the combination of NCTD,curcumin and adriamycin.Conclusions：The three kinds of Chinese herbs monomer have the effect on the induction of leukemia cell apoptosis and the enhancement effect of the NCTD and curcumin is related to the promoting effect of the intermalization adriamycin.
