乳房浸润性导管癌组织miR-10b和miR-195表达及意义

摘　　要：目的探讨miRNA 10b(miR-10b)、miRNA 195(miR-195)在乳腺浸润性导管癌组织表达及其意义。方法收集40例乳癌病人癌组织及癌旁组织标本,应用逆转录PCR法测定组织中miR-10b、miR-195表达,并分析其与临床病理特征的关系。结果乳腺浸润性导管癌组织miR-10b、miR-195表达明显低于癌旁组织(t=4.35、5.71,P<0.05)。miR-195表达与肿瘤的大小、有无淋巴结转移及间质脉管癌栓等有关(U=-2.127^-2.083,P<0.05),miR-10b、miR-195表达与肿瘤TNM分期、雌激素受体(ER)、孕激素受体(PR)及人表皮生长因子受体-2(Her-2)表达水平无明显相关性(P>0.05)。结论乳房组织中miR-10b、miR-195表达可能为筛查和诊断乳房肿瘤的潜在生物学指标。Objective To investigate the expression and its significance of miRNA 10b（miR-10b）and miRNA 195（miR-195）in tissues of patients with breast invasive ductal carcinoma. Methods Samples of cancer and paraneoplastic tissues of 40 breast cancer patients were collected,and the expression of miR-10 band miR-195 in the tissues was detected by using reverse transcriptase PCR,the relationship between the expression and clinicopathology was analyzed. Results The expressions of miR-10 b and miR-195 in the cancer tissue were significantly lower than that in the paraneoplastic tissue（t=4.35,5.71;P〈0.05）,of which,the expression of miR-195 was associated with the size of the cancer,with or without lymph node metastasis,or intravascular cancer emboli（U=-2.127--2.083,P〈0.05）.The expressions of miR-10 band miR-195 were not correlated with TNM stage,estrogen,progestogen,and Her-2（P〉0.05）. Conclusion The expressions of miR-10 band miR-195 in tissue of breast cancer are expected to become potential biological parameters in screening and diagnosing breast neoplasms.
