结直肠癌组织NEDD9的表达及临床意义

摘　　要：目的探讨神经前体细胞发育下调表达蛋白（NEDD9）在结直肠癌组织中表达及意义。方法采用免疫组织化学法,检测NEDD9在92例结直肠癌组织及其癌旁正常组织中的表达,并分析NEDD9表达与结直肠癌临床病理因素及预后的关系。结果 NEDD9在结直肠癌组织中表达阳性68例,癌旁正常组织中表达阳性12例,差异有显著性（χ2=65.35,P〈0.01）;NEDD9在结直肠癌组织表达与TNM分期及其中的pT、pN、pM有关（χ2=3.960-11.476,P〈0.05）。单因素和多因素生存分析显示,NEDD9高表达病人5年总生存率低于35%,NEDD9是影响结直肠癌预后的独立因素（P=0.025）。结论 NEDD9与结直肠癌的侵袭与转移密切相关,NEDD9高表达有可能成为判断结直肠癌病人预后的一个标记物,并为治疗提供新的分子靶点。Objective To investigate the expression of NEDD9 protein in colorectal cancer（CRC）and its significance.Methods With immunohistochemistry method,expression of NEDD9 in tissue of 92 cases of CRC and paraneoplastic normal tissue was measured,and the association of the expression with clinicopathology and prognosis was analyzed as well. Results Positive expression of NEDD9 was noted in 68 cases of CRC samples,that in paraneoplastic was 12 cases,the difference between them was significant（χ2=65.35,P〈0.01）.The expression of NEDD9 in CRC was associated with pT,pN and pM in TNM staging（χ2=3.960-11.476,P〈0.05）.Univariate and multivariate analysis indicated the overall 5-year survival rate in the patients with high NEDD9 expression was lower than 35%,NEDD9 was the independent factor that affected the prognosis of this disease（P=0.025）. Conclusion NEDD9 is closely correlated with the invasion and metastasis of CRC.The high expression of NEDD9 might serve as a marker to predict the prognosis of patients with colorectal cancer and provide a new molecular target point for the therapy.
