摘 要:目的观察富自体CGF纤维蛋白膜对人牙龈成纤维细胞黏附及增殖的影响,探讨CGF纤维蛋白膜作为生物膜和组织工程支架材料的可能性。方法于河北医科大学口腔医院口腔颌面外科门诊切取人正常牙龈组织,组织块贴壁培养法原代培养,鼠抗人波形蛋白、角蛋白鉴定。取第3代人牙龈成纤维细胞分别接种于富自体CGF纤维蛋白膜及Bio-Gide膜上培养。接种培养2天、5天MTT法测定人牙龈成纤维细胞增殖,接种培养5天后HE染色组织学观察、扫描电镜观察。结果人牙龈成纤维细胞在两种膜上均能黏附、增殖;接种培养第5天,人牙龈成纤维细胞在富自体CGF纤维蛋白膜上较Bio-Gide膜增殖明显(P<0.05)。结论富自体CGF纤维蛋白膜有利于人牙龈成纤维细胞黏附,能明显促进其增殖,有望成为组织缺损修复的生物膜及组织工程支架材料。Objective To observe the effect of the Concentrated Growth Fators(CGF) membrane on the proliferation and adhesion of human gingival fibroblasts, and to investigate the possibility of CGF membrane as a bioligical membrane and scaffold material for tissue engineering. Methods Normal human gingival tissue was obtained from patients with ages from 18 to 25 years in Department of Oral Maxillofacial Surgery, Hospital of Stomatology, Hebei Medical University. The fibroblasts were cultured by using traditional method of primary culture in vitro, the vimentin and cytokeratin antibodies were applied to identify the fibroblasts by Immunohistochemistry. The third generation of human gingival fibroblasts were respectively inoculated on CGF membrane and Bio-Gide membrane. The mean absorbance values of three groups were obtained by MTT assay after 2 days and 5 days of culture, after 5 days of culture, the cells of three groups stained with HE and Scanning electron microscope. Results Human gingival fibroblasts could grow and proliferate on two membranes; After 5 days of culture, there were significant difference between CGF membrane and BioGide membrane(P〈0.05). Conclusion CGF membrane has advantage to the adhesion of Human gingival fibroblasts,can obviously promote the proliferation of Human gingival fibroblasts, and is expected to become a kind of bioligical membrane and scaffold material for the reconstruction of tissue engineering.
