血清高迁移率族蛋白B1在非小细胞肺癌中的表达及其临床意义

摘　　要：目的:探讨血清高迁移率族蛋白B1(high mobility group box 1,HMGB1)在非小细胞肺癌(NSCLC)中的表达及其与肺癌临床、病理之间的关系和意义。方法:应用RT-PCR及Western-blot印记法检测30例经病理证实的非小细胞肺癌患者及15名健康对照组血清HMGB1水平,并分析与临床病理资料的关系。结果:非小细胞肺癌组血清HMGB1 mRNA及蛋白水平明显高于健康对照组。在非小细胞肺癌中,HMGB1的表达与TNM分期和细胞分化程度密切相关(P均<0.05),但与吸烟、性别、病理类型无关(P均>0.05)。高、中、低分化肺癌中HMGB1蛋白的表达量分别为(0.43±0.10)、(0.75±0.31)、(0.98±0.24),三者间差异具有统计学意义(P<0.05)。Ⅰ、Ⅱ期病例中HMGB1蛋白的表达量为(0.62±0.29),明显低于Ⅲ、Ⅳ期(0.85±0.32)(P<0.05)。结论:HMGB1在非小细胞肺癌中高表达,与肺癌TNM分期及细胞分化有关,可能作为判断非小细胞肺癌进展和预后判断的一种有效指标。To investigate the expression of HMGB1 in non - small cell lung cancer and to analyze the correlation between HMGB1 and clinicopathologic outcomes. Methods：The serum HMGB1 levels in 30 patients with non - smal cell lung cancer and 15 healthy controls were determined by real time PCR and Western - blot and analys-ize their relationship with clinical pathology parameter. Results：The expression of HMGB1 mRNA and protein was significantly higher in non - small cell lung cancer group than in healthy control group. Furthermore,expression of HMGB1 protein in non - smal cell lung cancer were related to TNM staing（P ﹤ 0. 05）and lung cancer cell differenti-ation（P ﹤ 0. 05）,but not to smoking,sex and histological type（P ﹥ 0. 05）. Multiple comparisons showed that there were different expressions of HMGB1 with different differentiation,（0. 43 ± 0. 10）,（0. 75 ± 0. 31）,（0. 98 ± 0. 24） （P ﹤ 0. 05）. HMGB1 protein of stage Ⅰ - Ⅱ（0. 62 ± 0. 29）were significantly higher than those of stage Ⅲ - Ⅳ（0. 85 ± 0. 32）（P ﹤ 0. 05）. Conclusion：HMGB1 may be a useful clinical marker for evaluating the NSCLC progres-sion and is of potential prognostic value.
