摘 要:目的观察32P-磷酸铬-聚L-乳酸(32 P-CP-PLLA)粒子瘤体植入后对荷H22肝癌KM小鼠移植瘤的抑瘤效果和对Caspase-3和Caspase-8蛋白表达的影响。方法建立荷H22肝癌KM模型小鼠45只后均分为粒子组(瘤内植入活度为7.4MBq的32P-CP-PLLA粒子1枚)、胶体组(瘤内注射活度为7.4MBq的32P-CP胶体)和对照组三组。分别于第1、6、12、24小时每组各处死3只小鼠,用分光光度法检测瘤体Caspase-3和Caspase-8的表达。每组3只小鼠每2天测量肿瘤体积,第14天测量肿瘤质量,计算抑瘤率。结果治疗第14天,粒子组抑瘤率高于胶体组[(62.28±2.36)%vs.(50.28±3.22)%](P<0.05)。与对照组比较,胶体组和粒子组各时间点的Caspase-3和Caspase-8的表达增高(P<0.05)。结论 32P-CP-PLLA粒子对荷H22小鼠移植瘤具有抑瘤和诱导细胞凋亡作用,适合于肝癌实体瘤的近距离治疗。Objective To investigate the effects of 32 P-chromic-poly L-lactide(s2 P-CP-PLLA) brachytherapy on H22 liver cancer cell-induced xenograft and the expressions of Caspase-3 and Caspase-8 in mice. Methods The xenograft models were established by transplantation of H22 tumor cells in 45 mice, which were equally assigned into three groups of A(32 P-CP-PLLA), B(3z P-chromic phosphate colloid) and C(model control). Three mice from each group were killed at 1,6, 12 and 24h, respectively, for detecting the expressions of Caspase-3 and Caspase-8 in tumor by spectrophotometry. The volume of xenograft in 3 mice from each group was measured every other day and tumor mass was detected on the 14th day, by which the inhibition rate was calculated. Results The inhibition rate on the 14th day was higher in group A than that in group BE(62.28±2.36)% vs. (50. 28±3.22)%](P〈0. 05). The expressions of Caspase-3 and Caspase-8 were higher in groups of A and B than those in group C(P〈0. 05). Conclusion 3z P-CP-PLLA is able to supress the growth and induce tumor cell apoptosis, which as brachytherapy is suitable to treat hepatocelluar carcinoma soild tumors.
