摘 要:目的探讨星形细胞瘤患者星形细胞上调基因-1(AEG-1)表达规律及其与星形细胞肿瘤恶性程度、预后之间的关系。方法随机抽取梅州市人民医院神经外科2000年1月至2012年6月间收治的星形细胞瘤患者80例(其中不同分化级别的样本各20例)。应用EnVision免疫组织化学染色法,检测所抽取病例中星形细胞瘤组织和正常脑组织中AEG-1的表达。比较不同级别星形细胞瘤的AEG-1表达水平及生存时间,分析AEG-1水平与星形细胞瘤分级及预后的关系。结果星形细胞瘤组织AEG-1总阳性率为95%(76/80),而正常脑组织中无AEG-1蛋白的特异性染色。不同级别肿瘤AEG-1阳性率随肿瘤恶性程度的增加而增高,表达强度与肿瘤的级别正相关(rs=0.481,P<0.01),与生存率呈负相关(rs=0.493,P<0.01)。结论 AEG-1表达水平和强度可作为判断星形细胞瘤分化程度和预后的生物学指标。Objective To discuss the relationship between the expression of astrocyte elevated gene- 1 ( AEG-1 ) and the severity and prognosis of the astrocytoma. Methods Clinical records from eighty astrocy- toma patients were randomly retrieved and retrospectively analyzed by hospital notes in the Department of Neurosurgery Meizhou People's Hospital from January, 2000 to June, 2012 (20 samples for each grade of astrocytoma). The expression level of AEG-1 in the astrocytoma and normal brain samples were studied by the EnVision immunohistochemical staining. The AEG-1 expression and survival rate of astrocytoma of varying grades were statistically compared. The association between AEG-1 expression and the grading and prognosis of astrocytoma was analyzed. Results In 80 tumor samples, 76 (95%) expressed AEG-1, while there was no AEG-1 expression in normal brain tissue. The positive rate of AEG-1 was increased with the elevating malignant grade of astrocytoma. The expression level of AEG-1 was positively associated with the malignant degree of astrocytoma ( ra = 0. 481, P 〈 0. 01 ), and negatively associated with survival rate ( ra = 0. 493, P 〈 0. 01 ). Conclusions The expression level and intensity of AEG-1 could serve as a biological index evaluating the differentiated degree and clinical prognosis of astrocytoma.
