摘  要:以河北大学食药用真菌研究所保藏的猴头(Hericium erinaceus)He001菌株为试材,对其进行原生质体紫外诱变处理,共获得273株再生菌株,初筛获得14株再生菌株与出发菌株有明显拮抗现象,进一步筛选出8株再生菌株菌丝长速优于出发菌株,并通过酯酶同工酶电泳技术对该8株菌株进行菌种真实性鉴定。结果表明:8株菌株共检测到4谱带,分布于迁移率在0.41-0.59之间;H035与出发菌株的联合系数为0.33,与He001遗传差异最大;同时它在栽培常用培养基上长速优于出发菌株,差异达到了极显著水平,因此初步认定其为优良的再生变异菌株。Use Hericium erinaceus He001 that stored in Institute of Medicinal Fungi of Heibei University,273 strains were regenerated in the process of protoplasts of He001 being exposed to the UV radiation. 14 strains were filtered out based on the mycelial antagonistic symbols,and 8 strains,of which mycelial growth rates were higher than the original,were chosen to detect their esterase isozyme to confirm whether they were genetically different from the original. The results showed that there were four bands of esterase isozyme located in the range of the migration rate between 0. 41 and 0. 59 in all. The association coefficient between H035 and He001 was 0. 33, which was the smallest and meant 11035 was genetically different from He001. Besides, the mycelia growth rate of strain H035 was higher than He001, and the difference was extremely significant. On the basis of all facts above, H035 was confirmed to be a mutant strain.
