血清VEGF、TGF-β_1及内皮抑素在非小细胞肺癌中的表达及相关性研究

摘　　要：目的检测血清中血管内皮生长因子VEGF、转化生长因子TGF-β1及内皮抑素(Endostatin)在非小细胞肺癌(NSCLC)中的表达情况。探讨三者与NSCLC临床病理特征间的关系及三者彼此的关系。方法用双抗体夹心酶联免疫吸附法ELISA检测63例非小细胞肺癌患者术前和28例健康体检者VEGF、TGF-β1及血清内皮抑素的水平。比较两组血清VEGF、TGF-β1及内皮抑素表达差异相关性及与临床病理特征的关系。结果①VEGF、TGF-β1及Endostatin血清中的阳性表达明显高于健康对照组(P<0.05);②血清VEGF、TGF-β1及Endostatin水平与NSCLC细胞分化程度、TNM分期、有无转移等密切相关(P<0.05),与组织学类型、有无淋巴结转移、患者年龄、性别和吸烟等无关(P>0.05);③NSCLC患者血清TGF-β1与VEGF水平呈正相关(P<0.01),Endostatin与VEGF及TGF-β1水平呈负相关(均P<0.05),是肿瘤血管形成重要的促进因子和抑制因子。结论 VEGF、TGF-β1及Endostatin共同参与了NSCLC血管生成,与细胞分化不良有密切关系,在非小细胞肺癌的发生、发展、转移中起重要作用,可能是预测肺癌恶性行为的有用指标。【Objective】To examine the serum levels of VEGF, TGF-β1, Endostatin in NSCLC and to study their relationship.【Methods】Serum levels of VEGF, TGF-β1, Endostatin were detected by ELISA in 63 patients with NSCLC before surgery and 28 healthy volunteers. difference correlation and the relationship with clinical pathological features in the serum levels of VEGF, TGF- β1and endostatin of two groups were compared. 【Results】①VEGF, TGF-β1and Endostatin in the serum of positive expression were higher than that of healthy controls（P 0.05）. ②The serum levels of VEGF, TGF-β1and Endostatin were closely correlated with cell differentiation, TNM stage, metastasis（P 0.05）, but were not correlated with histological type, lymph node metastasis, patient age, gender and smoking（P 0.05）. ③There were significantly positive association between TGF-β1and VEGF levels in serum of NSCLC patients（P 0.01）, and significantly negatively associated between Endostatin and TGF-β1, between Endostatin and VEGF（P 0.05）. They are important factors to promote tumor angiogenesis and inhibiting.【Conclusion】VEGF, TGF-β1and endostatin are closely related with NSCLC angiogenesis and poor cell differentiation, and promote the development and/or transfer of NSCLC. VEGF, TGF-β1and endostatin may be useful indicators for prediction of lung cancer.
