摘  要:目的研究锰超氧化物歧化酶(MnSOD)转染间充质干细胞(MSC)发挥抗自由基的细胞保护作用,并分析其机制。方法利用pcDNA3.1-MnSOD质粒转染MSC作为MnSOD-MSC实验组,仅转染pcDNA3.1为空白质粒组,未转染者为MSC对照组,在正常或高糖(含250 mmoL葡萄糖)培养条件下,MTT法观察MSC增殖情况,使用试剂盒检测各组MSC中谷胱甘肽氧化还原酶(GSHPX)、超氧化物歧化酶(SOD)表达情况。结果高糖环境下,与MSC对照组相比,MnSOD-MSC实验组MSC增殖OD值增高(P<0.01);与空白质粒组GSHPX相比,MnSOD-MSC实验组GSHPX含量明显增高(P<0.01),MnSOD-MSC实验组SOD含量在高糖条件下比空白质粒组增高(P<0.01)。结论 MnSOD转染MSC可以促进抗氧化酶类表达增高,对抗自由基,对MSC具有保护作用。[Objectives] To research cell protective effects against free radicals of mesenchymal stem cells (MSC) transfeeted by manganese superoxide dismutase (MnSOD), and to analyze its mechanism. [Metb- otis] pcDNA3.1-MnSOD plasmid transfecting MSC was used as MnSOD-MSC experimental group, peDNA3.1 transfecting only as blank plasmidgroup, untransfected as the MSC control group. Under normal or high sugar (including 250 mmol glucose) cultivating conditions, MTF method was used to observe the MSC prolifera- tion, and expression of glutathione oxidoreductase (GSHPX) and superoxide dismutase (SOD) in MSC of each group was detected by kits. [Results] Under high sugar environment, compared with MSC control group, in the MnSOD-MSC experimental group, OD value of MSC proliferation increased, with P 〈0.01; compared with the blank plasmid group GSHPX, in the MnSOD-MSC experimental group, GSHPX content increased obviously, with P〈0.01; in the MnSOD-MSC experimental group, under high sugar environment, content of SOD was higher than the content of SOD in the blank plasmid group, with P〈0.01. [Conclusion] MnSOD transfecting MSC can promote increase of antioxidant enzyme expression, with the function of fighting free radicals and protecting MSC.
