摘 要:目的 探讨叉状头/翅膀状螺旋转录因子(Foxp3)在食管癌细胞中的表达及临床意义.方法 采用免疫组织化学法检测103例食管癌患者肿瘤组织中Foxp3蛋白的表达,同时选用5例癌旁组织作为对照.通过对食管癌肿瘤细胞中Foxp3蛋白的表达水平进行H-score评分,并分析其临床意义.结果 免疫组织化学检测结果显示Foxp3蛋白在肿瘤浸润淋巴细胞及肿瘤细胞中主要表达在胞核,在癌旁组织中不表达;统计学分析显示,肿瘤细胞中Foxp3高表达(H-score≥70分)的患者中,肿瘤直径≥3.5 cm患者所占比率显著高于肿瘤细胞中Foxp3低表达(H-score< 70分)的患者,差异有统计学意义(x2 =4.267,P <0.05),而肿瘤细胞中Foxp3表达水平与患者其他临床参数无相关(P>0.05).结论 核转录因子Foxp3在食管癌组织中表达主要分布在浸润淋巴细胞及肿瘤细胞,肿瘤细胞表达Foxp3在肿瘤进展中有重要意义.Objective To investigate the expression of forkhead box P3 (Foxp3) in human esophageal cancer cells and its clinical significance.Methods Immunohistochemistry assay was used to detect the Foxp3 protein expression in 103 cases of esophageal cancer tissues and 5 cases of adjacent normal tissues,and H-score method was used to assess Foxp3 expression level in esophageal cancer cells and to analyze its clinical significance.Results Immunohistochemistry results showed that the Foxp3 protein predominantly localized on the nuclei of tumor infiltrating lymphocytes as well as esophageal cancer cells,and was negatively expressed on adjacent normal tissues.Based on the H-score evaluation of Foxp3 protein immunostaining,we found that in patients with higher Foxp3 expression (H-score≥70),the ratio of tumor size ≥3.5 cm patients was significantly higher than that in patients with lower Foxp3 expression (H-score < 70) (x2 =4.267,P < 0.05),while the Foxp3 expression level in tumor cells was not correlated to other clinical parameters.Conclusion The nuclear transcription factor Foxp3 expression could be found in cancer cells and tumor infiltrating lymphocytes in human esophageal cancer,and the cancer cell expressed Foxp3 protein may play an important role in cancer progression,and could be used as a potential target in the diagnosis and therapeutics of esophageal cancer.
