摘 要:目的 探讨糖原合成激酶-3β(GSK-3β)在胰腺癌组织中的表达及其与上皮-间充质转化的相关性.方法 选取胰腺癌病例标本60例,35例有配对的癌旁正常组织,行GSK-3β、丝氨酸-9、酪氨酸-216、Snail和E-钙黏蛋白(E-cadherin)免疫组织化学及Western blot检测,分析GSK-3β与胰腺癌临床病理特征和上皮-间充质转化相关分子之间的相关性.结果 GSK-3β在胰腺癌中阳性率为65.0%,显著高于癌旁组织(22.8%),GSK-3β表达与胰腺癌临床分期、分级、分化程度以及淋巴结转移呈正相关,而与患者年龄、性别和肿瘤大小无相关.丝氨酸-9在癌旁组织表达水平高于胰腺癌组织,而酪氨酸-216和Snail表达与之相反,E-cadherin在胰腺癌及癌旁组织表达水平均较低.Pearson相关分析发现,GSK-3β与上述指标均显著相关.结论 GSK-3β在胰腺癌组织中表达显著高于癌旁组织,与胰腺癌临床分期、肿瘤分期、分化程度及淋巴结转移之间呈正相关,而与患者年龄、性别及肿瘤大小无相关.此外,GSK-3β还与胰腺癌上皮-间充质转化具有密切关系.Objective To explore the glycogen synthase kinase-3β (GSK-3β) expression in pancreatic carcinoma and its correlation with epithelial-mesenchymal transition.Methods Sixty cases of pancreatic adenocarcinoma specimens dated from 2006-2010 were adopted in this study,35 of which had paired adjacent normal tissues.Immunohistochemical staining and Western blotting were used to detect the expression of GSK-3β,Ser-9,Tyr-216,Sanil and E-cadherin.The correlation between GSK-3 β and pancreatic cancer clinicopathological parameters,or epithelial-mesenchymal transition was analyzed.Results The positive expression rate of GSK-3 β in pancreatic carcinoma was 65.0%,which was significantly higher than in paired adjacent normal tissues (22.8%),and GSK-3β expression was positive correlated with pancreatic cancer clinical stage classification,degree of differentiation and lymph node metastasis (P < 0.05),but not with patient's age,gender and tumor size (P > 0.05).Phosphorylation of GSK-3 β (Ser-9) in the paired adjacent normal tissues was significantly higher than in pancreatic cancer,and the expression of Tyr-216 and Snail was quite the opposite.E-cadherin expression was lower in both cancer and paired adjacent normal tissues.Pearson correlation analysis revealed that GSK-3β was correlated with other abovementioned markers.Conclusion GSK-3β expression was significantly higher in pancreatic cancer than in the paird adjacent normal tissues,and had a positive correlation with clinical stage,TNM stage,tumor differentiation and lymph node metastasis.In addition,GSK-3β had significant relationship with epithelialmesenchymal transition.
