家蚕追寄蝇寄生蚕体的血液蛋白质差异表达分析及质谱鉴定  被引量:2

摘  要:由家蚕追寄蝇(Exorista sorbillans)寄生于家蚕幼虫体壁与肌肉间引起的多化性蚕蛆病对蚕业生产危害极大。基于家蚕血液在蚕体基础代谢、生长发育及先天免疫中的重要功能,采用双向电泳(2-DE)技术检测被家蚕追寄蝇寄生的家蚕5龄幼虫至蛹期第2天的血液蛋白质表达变化,并对表达水平具有显著差异的蛋白点进行质谱鉴定,明确家蚕在受到追寄蝇感染时与免疫应答以及生理、病理变化过程有关的血液蛋白质。在被追寄蝇寄生的蚕体血液中共检测到24个具有显著表达差异的蛋白点,表达上调的蛋白点有17个,包括在家蚕先天免疫系统中发挥重要作用的肽聚糖识别蛋白-S1、C-型凝集素-10和酚氧化酶-2等;表达下调的蛋白点有7个,包括与家蚕生长发育、体内物质代谢等生物学过程相关的蛋白质。血液蛋白质的表达变化提示,当蚕体受到追寄蝇感染时其先天免疫系统被激活,生长发育和体内物质代谢等也受到影响。The multivoltine maggoty disease caused by parasitism of Exorista sorbillans Wiedemann between the epidermis and the muscle of silkworm(Bombyx mori) larvae leads to great harm to sericultural production.Based on the crucial functions of silkworm haemolymph in basal metabolism,growth,development and innate immune system,silkworm haemolymph proteins from the 2nd day pupae developed from the 5th instar larvae which were parasitized by Exorista sorbillans were analyzed using two-dimensional electrophoresis(2-DE) technology.The protein spots with significantly different expression levels were identified by mass spectrometry,and the haemolymph proteins involved in immune response,physiological and pathological changes after parasitism of Exorista sorbillans were clarified.Totally 24 protein spots with obvious expressional difference were identified in silkworm haemolymph after parasitism of Exorista sorbillans.Among them,17 protein spots were up-regulated,including peptidoglycan recognition protein S1,Ctype lectin 10 and Phenoloxidase 2,all of which play important roles in the innate immune system.Seven protein spots were down-regulated,including some proteins relevant to growth,development,metabolism and other bio-logical processes.These results indicate that the innate immune system was activated while the silkworm was infected by Exorista sorbillans,and growth,development and metabolism of the silkworm were also affected.
