摘  要:桑树的乳汁采集自桑枝叶腋处,其中含有较丰富的生物碱类物质1-脱氧野尻霉素(DNJ)。采集不同桑品种及不同生长季节、生长部位桑枝叶腋处的乳汁,用柱前衍生化反相高效液相色谱-紫外检测法测定乳汁中的DNJ含量,并分析乳汁样品及其提取液在不同温度条件下贮存的DNJ稳定性以及桑树乳汁中DNJ含量的日变化规律。不同桑品种及不同倍数性品种、不同生长季节及不同部位桑树乳汁中的DNJ含量有较大差异:供试品种均是7~8月份采集桑树乳汁中的DNJ含量最高,其中二倍体丰产型品种新一之高达993μg/mL,明显高于其诱变培育的三倍体品种陕桑305,比耐旱型二倍体品种吴堡桑高出6.7倍(P〈0.05);在供试品种陕桑305中部枝条叶腋采集桑树乳汁中的DNJ含量相对较高,可达811μg/mL,与上部枝条和下部枝条相比差异极显著(P〈0.01)。桑树乳汁在常温、4℃和-20℃条件下短时间(28 d内)保存的DNJ含量相对稳定,其粗提液中的DNJ则容易降解。丰产型品种新一之和陕桑305桑树乳汁中的DNJ含量均呈现相同的日变化规律。研究结果提示:桑品种及其倍数性,桑树的生长季节及光照强度,桑树植株的部位等都对桑树乳汁中的DNJ含量有影响;桑树乳汁直接在适宜的温度下短期贮存可以减少DNJ的降解。Mulberry latex can be collected from leaf axil of mulberry branch.Mulberry latex contains abundant 1-deoxynojirimycin(DNJ) which is one of the alkaloid substances.Mulberry latex samples were collected from leaf axil of mulberry branch of different varieties,different growth stages and different positions.Then,pre-column derivatization reverse phase high performance liquid chromatography and ultraviolet detection method were employed to determine DNJ content in mulberry latex.Furthermore,stability of DNJ content in latex samples and in extraction solution stored under different conditions and diurnal variation of DNJ content in mulberry latex were analyzed.The results indicated that DNJ content in mulberry latex was greatly varied with mulberry species,ploidy of mulberry variety,growth stage,and collection position.All tested mulberry varieties had the highest DNJ content in latex collected from July to August,among which diploid high-yield variety Shin Ichinose reached 993μg /mL,which was apparently higher than triploid Shansang 305 and was 6.7 times higher than diploid drought resistant variety Wubaosang(P 〈 0.05).Latex from leaf axil at middle position of mulberry branch was relatively higher in mulberry variety Shansang 305(811 μg /mL),which had extremely significant difference with those collected at up and low positions of branch(P 〈0.01).DNJcontent was relatively stable in mulberry latex stored at normal temperature,4 ℃,and -20 ℃ for a short period(within28 days),while DNJ in crude extraction solution was degraded more easily.High-yield variety Shin Ichinose and Shansang 305 presented the same diurnal variation in DNJ content of latex.These results suggest that DNJ content is affected by mulberry variety and ploidy,growth stage,light intensity,and collection position,and short period storage of mulberry latex under suitable temperature can reduce degradation of DNJ.
