摘 要:目的探讨miR-124对紫杉醇诱导的乳腺癌细胞株MCF-7生长抑制作用的影响。方法应用MTr法检测紫杉醇对MCF-7的生长抑制作用,流式细胞仪检测紫杉醇对细胞周期的影响,应用实时荧光定量PCR(qRT-PCR)技术检测紫杉醇处理MCF-7细胞后,miR-124表达水平的变化,进一步转染miR-124抑制剂至MCF-7细胞中,用MTr法检测细胞生长抑制情况。结果MrI-T法检测结果显示紫杉醇能显著抑制MCF-7细胞生长。流式细胞仪检测结果显示紫杉醇能明显将细胞阻滞于G:期。qRT-PCR检测结果显示紫杉醇能诱导miR-124表达水平升高,并呈现剂量依赖关系。进一步研究发现,抑制细胞miR-124的表达后,紫杉醇对MCF-7的增殖抑制作用受到显著影响。结论紫杉醇能显著诱导MCF-7上调表达miR-124,并且这种上调表达是剂量依赖性的。而miR-124抑制剂能明显降低紫杉醇对MCF-7细胞的生长抑制作用。表明miR-124可能在细胞对紫杉醇的化疗耐药中起着重要作用,也为将来解决紫杉醇临床耐药提供了新思路。Objective Our study aims to investigate the inhibitive effects of miR - 124 on the growth of breast cancer cell MCF - 7 induced by paehtaxel. Methods MTr was used to detect the growth inhibition of MCF -7 induced by paclitaxel. Flow cytometry was used to detect the effect of paclitaxel on cell cycle. Real - time quantitative PCR( qRT - PCR) was used to detect the expressive level of miR - 124, while MCF - 7 cells were treated with paclitaxel. MiR- 124 inhibitor was transfected into MCF-7 breast cancer cells,and growth in- hibition was detected by MTI'. Results The results showed that paclitaxel could significantly inhibit the growth of breast cancer cell line MCF -7 by blocking the G2 phase. The results from qRT - PCR showed that the relative expression of miR - 124 was increased when the dosage of paclitaxel was increased. When the expression of miR - 124 was inhibited, the cell growth inhibition caused by paclitaxel was also prominently decreased. Conclusion The higher expression of miR - 124 in MCF - 7 induced by paclitaxel was dose dependent. And miR - 124 in- hibitor can significantly influence the cell growth inhibition caused by paclitaxel. These results indicat that miR - 124 plays an important role in paclitaxel - induced chemotherapy drug resistance, and provides a new direction to solve the problem.
