摘 要:目的研究缺血后大鼠脑NOS阳性神经元的变化,阐明脑缺血对NOS表达的影响及其意义。方法30只成年雌性SD大鼠,随机分为正常组和缺血组,采用线栓法制备右侧大脑中动脉闭塞模型,24小时后常规灌注取脑,冰冻切片,用黄递酶法进行NOS染色,计数两组大鼠海马各区CA1、CA2、CA3、DG和皮质区NOS神经元的数目。结果正常大鼠海马各区及皮质区左右两侧NOS神经元数目无显著差异(P〉0.05);缺血组右侧CA1区NOS神经元数目高于左侧(P〈0.05),其余各区无显著差异(P〉0.05);与正常组比较,缺血组双侧CA1、CA3、DG区NOS神经元数目明显高于正常组(P〈0.05),CA2区和皮质区则无显著差异(P〉0.05)。结论缺血会导致海马NOS表达增加,NO生成增加,产生神经毒性,这可能缺血导致脑损伤的一个机制。调节NO表达可能是治疗脑缺血的一个方法。Objectve To study the alterations of nitric oxide synthase (NOS) positive neuron in rat brain after brain ischemia and to interpret the influence of ischemia on NOS and its clinical implications. Methods Thirty female adult SD rats were divided into normal and ischemia group randomly. The right middle cerebral artery occlusion (MCAO) model was made by suture-occluded method. Twenty-four hours later, the animals were sacrificed and brains were picked up. Frozen brain sections were stained by NADPH-d method. The numbers of NOS positive neurons in CA1, CA2, CA3, dentate gyrus and cortex were counted. Result The number of NOS positive neurons in each area of hippocampus and cortex of normal rates are no differences between left side and right side (P〉0.05); the number of NOS positive neurons in the right CA1 area is higher than that of left (P〈O.05), but no difference in other areas(P〉0.05); the number of NO$ positive neurons in the right CA1, CA3 and dentate gyrus of the ischemia rats are higher than that of normal group (P〈O.05), but no difference was found in CA2 and cortex(P〉0.05). Conclusions Ischemia can result in increasing NO$, which contributed to increasing nitric oxide production, and lead to neurotoxicity. This may be a mechanism of brain injury induced by brain ischemia. Adjust nitric oxide expression maybe a method to cure brain ischemia.
